猫冠状病毒的临床流行特征及其遗传进化分析

【摘要】 为了解当前猫冠状病毒(Feline coronavirous,FCoV)的流行情况,本研究在2018-2020年期间收集来自广西、河南、广州和江西等地疑似猫传染性腹膜炎(Feline infectious peritonitis,FIP)病例,共计108份。通过RT-nPCR方法对样品进行鉴定和部分S基因扩增和遗传进化分析。结果显示,108份样品中阳性样品共计66份,阳性率高达61.1%,血清型分型均为Ⅰ型。将扩增所获得的57条S基因的分型序列与国内外其他参考毒株序列进行遗传进化分析,结果发现FCoVs具有高度的遗传多样性,其中55个毒株与欧洲、日本和中国毒株都分别在不同分支,它们之间的核苷酸同源性为81.8~93.7%。值得关注的是,来自同一只猫的不同脏器毒株(FCoV GXBH-10)被划分在两个分支中,其中肺脏毒株与美国毒株OH11927亲缘关系较近,核苷酸同源性为93.8%,而其他脏器毒株却在另一个分支,暗示了共感染现象。此外,本研究对S蛋白3’末端19条氨基酸序列进行分析,结果发现突变位置主要发生在3个区域,分别为D1267H1268→EY/EQ、H1444→N和T1454→N/S,是否这些突变可作为遗传标记,有待进一步验证。本研究针对近年来FCoV的流行趋势进行了系统分析,将为今后掌握FCoV流行规律及早期临床诊断提供重要的参考依据。

【Abstract】 In order to understand the prevalence of feline conronavirus(FCoV),total of 108 samples were collected from suspected feline infectious peritonitis(FIP) cases were collected from Guangxi,Henan,Guangzhou and Jiangxi between 2018 and 2020.The samples were identified by RT-nPCR and partial S genes were amplified,the phylogenetic tree was constructed and the sequences of amino acids were analyzed.The results showed that 66 FCoV-positive samples were detected in 108 clinical samples.The positive rate was as high as 61.1%.The typing of serotype was identified for type Ⅰ.The typing sequences of 57 S genes obtained were performed the analysis of genetic evolution,compared with other reference strains at home and abroad,the results showed that FCoVs owned high genetic diversity,among which 55 FCoV strains were divided in different branches compared with European,Japanese and Chinese strains,and their nucleotide homology was 83.5~100%.Notably,different organ strains(FCoV GXBH-10) from the same cat were grouped with two branches,among which the lung strain was closely related to the American strain OH11927,with a nucleoside homology of 93.5%,while other organ strains fell into another branch,suggesting the phenomenon of co-infection.In addition,the 19 amino acid sequences of the 3’terminal of S protein were analyzed,it was found that the mutations were located mainly in three regions,namely D1267H1268→EY/EQ,H1444→N andT1454→N/S,whether these mutations can be used as genetic markers remains to be further verified.In this study,the epidemic trend of FCoV in recent years was systematically analyzed,which will provide important reference for understanding the epidemic pattern and early clinical diagnosis of FCoV.
