百年兴衰沉浮:食品生物大分子复合凝聚与液-液相分离
A century of ups and downs:complex coacervation of food biomacromolecules and liquid-liquid phase separation

【摘要】 复合凝聚是一种由大分子间多价相互作用引起的去溶剂化和液-液相分离过程。这一现象于1911年被德国化学家F.W.Tiebackx首次报道,并在1929年由荷兰化学家Bungenberg-de Jong和H.R.Kruyt进行了命名和系统研究。由于复合凝聚与原型细胞及生命过程的潜在联系,相关研究在百余年来经历了从兴盛到衰落,再到复兴的发展历程。2004年,Russell J.Stewart发现一些海洋无脊椎动物(如沙堡蠕虫)利用复合凝聚机制合成耐水生物粘合剂,促使了对相关机制和复合凝聚型水下粘附材料长达20年的研究探索。2009年,Tony Hyman团队和Michael Rosen团队几乎同时独立发现无膜细胞器是蛋白质和核酸等大分子液-液相分离的杰出作品,这一发现催生了十五年来蓬勃发展的生物分子凝聚体和仿生细胞领域,并重新唤醒了沉寂多年的凝聚物研究。蛋白质和多糖等食品生物大分子是最早被发现的复合凝聚体系,但相关研究长期停滞在现象学层面。Flory-Huggins和Voorn-Overbeek等百年历程中的理论明珠,以及近年来基于合成聚电解质复合凝聚的仿生学相态和多尺度动力学理论如何启发食品生物大分子复合凝聚的进一步发展引人深思。
【Abstract】 Complex coacervation is a desolvation and liquid-liquid phase separation process caused by multivalent interactions between macromolecules.This phenomenon was first reported by German chemist F.W.Tiebackx in 1911,and was named and systematically studied by Dutch chemists Bungenberg-de Jong and H.R.Kruyt in 1929.Due to the potential connection between complex coacervation and protocells and life processes,related research has experienced a development process from prosperity to decline and then revival over the past century.In 2004,Russell J.Stewart discovered that some marine invertebrates(such as sandcastle worms) use complex coacervation mechanisms to synthesize water-resistant bioadhesives,which prompted 20 years of research and exploration of related mechanisms and complex coacervation-type underwater adhesive materials.In 2009,Tony Hyman’s team and Michael Rosen’s team almost simultaneously independently discovered that membraneless organelles are outstanding works of liquid-liquid phase separation of macromolecules such as proteins and nucleic acids.This discovery gave rise to the field of biomolecular condensates and artificial cells,which has flourished for fifteen years,and reawakened the research on condensates/coacervation that had been dormant for many years.Food biomacromolecules such as proteins and polysaccharides are the earliest discovered complex coacervation systems,but related research has long stagnated at the phenomenological level.It is thought-provoking how the theoretical pearls of the century-long history such as Flory-Huggins and Voorn-Overbeek,as well as the biomimetic phase state and multi-scale dynamics theory based on synthetic polyelectrolyte complex coacervation in recent years,can inspire the further development of food biomacromolecule complex coacervation.
