黄菇娘致腐菌的分离与鉴定

【摘要】 为明确市场上黄菇娘致腐菌种类,延长黄菇娘的储藏期。以腐败变质的黄菇娘果为原料,通过多次稀释涂布平板法进行单菌落形态学观察,将菌株反接种到新鲜菇娘果上,筛选出腐败菌,以真菌ITS序列、细菌16S r DNA序列构建发育树,鉴定到属,再将鉴定结果与分离的菌株的形态学鉴定结果对比,验证菌株鉴定结果。结果表明:本研究从腐败变质的黄菇娘果中分离鉴定出3株典型致腐菌,分别为6号西毕赤酵母(Pichia occidentalis),16号肠膜明串珠菌(Leuconostoc mesenteroides),21号库氏毕赤酵母(Pichia kudriavzevii)。本研究结果有利于进一步深入研究致腐微生物及其对黄菇娘水果病害的致病机理,以便进一步控制果实的健康安全,并为下一步黄菇娘果实抗病提供一定参考,以期寻找到有效对抗致腐菌的生防菌株,为黄菇娘果保鲜提供更多支持,对后续黄菇娘果的采集、运输以及保鲜奠定基础。

【Abstract】 In order to clarify the species of spoilage-causing bacteria in the market and to prolong the storage period of yellow mushroom.We used spoiled yellow mushroom fruits as raw materials,observed the morphology of single colonies by multiple dilution smear plate method,back-inoculated the strains onto fresh mushroom fruits,screened out the spoilage bacteria,constructed developmental trees with fungal ITS sequences and bacterial 16S r DNA sequences,identified them to genera,and then verified the results of strain identification by comparing the results with those of the morphological identification of the isolated strains.The results showed that in this study,three strains of typical rot-causing bacteria were isolated and identified from rotting and spoiled yellow mushroom fruits,which were Pichia occidentalis(No.6),Leuconostoc mesenteroides(No.16),and Pichia kudriavzevii(No.21).The results of this study are conducive to further in-depth research on the rotcausing microorganisms and their pathogenic mechanisms on yellow mushroom fruit diseases,in order to further control the health and safety of the fruits,and to provide some reference for the next step of disease resistance of yellow mushroom fruits,with a view to finding biocontrol strains that are effective in fighting against the rot-causing fungi,and to provide more support for the preservation of the yellow mushroom fruits,and laying the foundation for the subsequent collection,transportation,and preservation of the fruits of yellow mushrooms.
