茶树查尔酮合酶基因CHS2单核苷酸多态性研究
Identification of Single Nucleotide Polymorphisms in Chalcone Synthase Gene(CHS2)in tea plant(Camellia sinensis)

【摘要】 以21个茶树品种为材料,根据茶树查尔酮合酶(chalcone synthase,CHS) CHS2基因cDNA序列(D26594)两端非编码区设计引物,进行PCR扩增,获得CHS2外显子Ⅰ和外显子Ⅱ全部序列。研究结果发现:(1)外显子Ⅰ有15个SNPs,2个InDels,单倍型多样性指数(Hd)为0.813,核苷酸多样性指数(Pi)为0.01207;(2)外显子Ⅱ有37个SNPs,28个InDels,单倍型多样性指数为0.962,核苷酸多样性指数为0.0505;(3)聚类分析结果表明,茶树CHS2基因外显子Ⅱ能较好反映不同茶树品种适制性。
【Abstract】 The exon Ⅰ and exon Ⅱ of Camellia sinesnsis gene CHS2 were amplified by designed primers according to non-coding sequences of CHS2(D26594) to identify the SNPs among 21 tea plant varieties.Results showed that:(1) In exon I,there were 15 SNPs and 2 Indels,and Haplotype Diversity Index(Hd) and Nucleotide Diversity Index(Pi) were 0.813 and 0.01207,respectively.(2) In exon Ⅱ,there were 37 SNPs and 28 Indels,and Haplotype diversity index(Hd) and Nucleotide diversity index(Pi) were 0.962 and 0.0505,respectively.(3) Cluster analysis indicated that exon Ⅱ of CHS2 could classify manufacture suitablility to some degree.
