薯蓣碳点对金黄色葡萄球菌的抑菌作用研究

【摘要】 目的:研究薯蓣碳点对金黄色葡萄球菌的抑菌作用,为薯蓣碳点对金黄色葡萄球菌感染的临床应用提供实验依据。方法:采用一步水热法合成薯蓣碳点。制备细菌涂片,通过革兰氏染色观察细菌形态,初步鉴定菌种。抑菌环实验判断薯蓣碳点对金黄色葡萄球菌的抑菌能力,设置分组:薯蓣碳点组(50mg/mL薯蓣碳点),阳性对照组(0.12%氯己定),阴性对照组(生理盐水)。通过倍量稀释法测定薯蓣碳点对金黄色葡萄球菌的最小抑菌浓度(MIC)。结果:革兰氏染色显示金黄色葡萄球菌革兰染色阳性,形态为球形,单个、成对或葡萄状排列,直径0.5-1.0um,无杂菌。抑菌环实验显示50mg/mL薯蓣碳点形成直径为12mm的抑菌环,比阳性对照组稍低,药敏结果为中度敏感。薯蓣碳点MIC为6.25mg/mL。结论:作为中药来源的一类新型的碳基纳米材料,薯蓣碳点对金黄色葡萄球菌良好的抑制作用,为开发绿色廉价且高效的新型抑菌材料提供新思路。

【Abstract】 OBJECTIVE:To study the antibacterial effect of Dioscorea carbonica dots on Staphylococcus aureus and to provide experimental basis for the clinical application of Dioscorea carbonica dots on Staphylococcus aureus infection.METHODS:Dioscore was synthesized by one-step hydrothermal method.Bacterial smears were prepared,and the morphology of the bacteria was observed by Gram staining to identify the bacterial species.The inhibitory ring test was performed to determine the inhibitory ability of Dioscorea carbonica dots against Staphylococcus aureus,and the groups were set up as follows:Dioscorea carbonica dots group(50mg/mL Dioscorea carbonica dots),positive control(0.12% chlorohexidine),and negative control(physiological saline).The minimum inhibitory concentration(MIC) of Dioscorea and Dioscorea carbonica dots against Staphylococcus aureus was determined by doubling dilution method.RESULTS:Gram stain showed positive Gram stain of Staphylococcus aureus with a spherical morphology,arranged singly,in pairs or grapelike,0.5-1.0 um in diameter and free of stray organisms.The ring of inhibition test showed that 50 mg/mL Dioscorea carbonica dots formed a ring of inhibition with a diameter of 12 mm,which was slightly lower than that of the positive control group,and the drug sensitivity result was moderately sensitive.The MIC of Dioscorea carbonica dots was 6.25mg/mL.CONCLUSION:As a new class of carbon-based nanomaterials derived from traditional Chinese medicine,Dioscorea carbonica dots showed good inhibitory effects against Staphylococcus aureus,providing new ideas for the development of new green,cheap and efficient bacteriostatic materials.
