嗜酸乳杆菌LA对大肠杆菌O157:H7体外抑菌作用及益生特性研究

【摘要】 本实验为了研究从牛乳中分离除的嗜酸乳杆菌LA(LA)的益生特性,利用牛津杯法测定LA上清液和灭活菌液对大肠杆菌O157:H7(EC)的抑菌能力,利用分光光度法对比LA和植物乳杆菌(LP)的自聚合能力以及与食源性致病菌共聚合能力,并研究了LA对人工模拟胃液、肠液、胆盐耐受性等基本益生特性。结果表明LA具有抑制EC的能力,其灭活菌液抑制EC的效果较为显著,抑菌圈直径为(20.47±0.43) mm,经过实验可以得出通过酶处理的菌液对EC抑制作用明显减少,抑菌圈直径为(5.68±0.34) mm,因此LA的抑菌物质初步确定为细菌素类物质。LA的自聚合率显著高于LP为27.51%,在共聚实验中,LA与EC的共聚能力显著高于LP,其共聚合率为24.23%。因此LA具有自聚作用和与致病菌共聚作用。此外通过LA对人工模拟胃肠液和胆盐的耐受性实验可以得出LA具有较好的胃肠道耐受性。

【Abstract】 This study to investigate the isolated from milk in addition to the live properties of Lactobacillus acidophilus LA(LA),Oxford cup method has been applied to the determination of LA on clear liquid and inactivated bacteria liquid for E.coli 0157:H7(EC) antibacterial ability,using spectrophotometric method contrast of LA and Lactobacillus plantarum(LP)since the abilities of polymerization and copolymerization of foodborne bacterial pathogens,The basic probiotic properties of LA on artificial simulated gastric juice,intestinal juice and bile salt tolerance were also studied.The results showed that LA had the ability to inhibit EC,and its inactivated liquid had a significant effect on EC,and the diameter of the inhibition zone was(20.47±0.43) mm.Through the experiment,it could be concluded that the inhibitory effect of LA on EC was significantly reduced.The diameter of the inhibition zone was(5.68±0.34) mm.Therefore,bacteriocins were initially determined as the antibacterial substances of LA.The self-polymerization rate of LA was significantly higher than that of LP(27.51%).In the copolymerization experiment,the copolymerization ability of LA and EC was significantly higher than that of LP(24.23%).Therefore,LA has self-polymerization and co-polymerization with pathogenic bacteria.In addition,the tolerance test of LA to artificial simulated gastric fluid and bile salts showed that LA had good gastrointestinal tolerance.
