麦角甾醇及其过氧化物的抗炎功效对比

【摘要】 麦角甾醇(Ergosterol),别名麦角固醇,是丝状真菌和酵母菌中的主要甾醇,对细胞膜和细胞内多种生物学过程具有重要的调控作用。本课题预实验发现其能够降低皮肤细胞炎症因子的分泌。由此,对麦角甾醇及其过氧化物在皮肤细胞上的抗炎功效进行对比研究。分别建立LPS和UVA诱导的人角质形成细胞(HaCaT)炎症模型,通过细胞活力测试,考察它们对炎症状态下HaCaT细胞的保护和修复能力;利用炎症因子人白细胞介素-8(IL-8)和人白细胞介素-1β(IL-1β)的表达情况及分析皮肤水通道蛋白3(AQP3)和丝聚蛋白(FLG)含量变化,评价它们的抗炎功效和对炎症损伤皮肤屏障相关功能的影响。结果表明:麦角甾醇及其过氧化物对炎症损伤的HaCaT细胞具有保护和修复作用;能够降低细胞内ROS水平;对IL-8和IL-1β炎性因子均有抑制作用;对AQP3和FLG具有显著的促进作用。其中,麦角甾醇的保护和修复能力较好。研究表明,麦角甾醇及其过氧化物对UVA和LPS诱导的炎症损伤有一定程度的保护和修复作用,为今后的开发应用提供了理论基础。

【Abstract】 Ergosterol is the main sterol in filamentous fungi and yeasts,and has important regulatory roles in cell membranes and a variety of intracellular biological processes.Pre-experiments in this subject revealed its ability to reduce the secretion of inflammatory factors in skin cells.This led to a comparative study of the anti-inflammatory efficacy of ergosterol and its peroxide on skin cells.Establishment of LPS and UVA-induced inflammation models of human keratinocytes(HaCaT) and to examine their protective and reparative abilities against HaCaT cells in the inflammatory state by cell viability tests.Using the expression of the inflammatory factors human interleukin-8(IL-8),human interleukin-1β(IL-1β) and the analysis of changes in the content of skin aquaporin 3(AQP3),filaggrin(FLG) and to evaluate their anti-inflammatory efficacy and effect on the function related to inflammatory damage to the skin barrier.The results showed that ergosterol and its peroxide had protective and reparative effects on inflammation-damaged HaCaT cells;reduced intracellular ROS levels;inhibited both IL-8 and IL-1β inflammatory factors;and had significant promotion effects on AQP3 and FLG.Among them,ergosterol has a better protective and repairing ability.The study showed that ergosterol and its peroxide have a certain degree of protective and repairing effect on UVA and LPS-induced inflammatory damage,which provides a theoretical basis for future development and application.
