绿豆肽螯合亚铁对小鼠缺铁性贫血改善作用研究

【摘要】 本研究旨在探究绿豆肽螯合亚铁(MBP-Fe)的体内补铁功效。对绿豆蛋白进行酶解后,经超滤、Sephadex G-15纯化分离出小分子绿豆肽,采用Nano RP-HPLC与Q Exactive Plus质谱联合鉴定其氨基酸序列,经UniProtKB蛋白库筛选比对得到6条平均分子量为825 u的肽,其中每个肽段都含有一个或多个酸性氨基酸,如Glu和Asp,以及碱性氨基酸,如Arg和Lys。通过动物实验确定MBP与Fe2+改善缺铁性贫血(IDA)的体内功效。ICR小鼠随机分为7组:1组为正常对照组,其余6组利用缺铁饲料建立IDA模型后设为1组缺铁模型组和5组缺铁组,缺铁组分别补充甘氨酸亚铁(3.5 mg Fe/kg BW)、MBP协同吸收Fe2+(MBP:FeSO4=8:1,3.5 mg Fe/kg BW),以及低、中、高剂量MBP-Fe(1.16,1.75,3.5 mg Fe/kg BW),灌胃28 d。结果发现高剂量MBP-Fe和MBP协同吸收Fe2+组(3.5 mg Fe/kg BW)小鼠血清铁、血红蛋白及体重增幅显著增加(P <0.05),并恢复到正常水平,脾脏系数显著降低,相比于甘氨酸亚铁组。综上,MPH-Fe及MPH协同Fe2+吸收在达到有效量后对IDA均具有显著的改善作用,而MPH-Fe效果更优。

【Abstract】 The purpose of this study was to explore the iron supplementation effect of iron chelating peptide from mung bean(MBP-Fe) in vivo.After enzymatic hydrolysis of mung bean protein,the small molecule mung bean peptides were separated by ultrafiltration and Sephadex G-15 purification.The amino acid sequences were identified by Nano RP-HPLC and Q Exactive Plus mass spectrometry.Six peptides with an average molecular weight of 825 u were obtained by screening and comparison of the UniProtKB protein library.Each peptide contained one or more acidic amino acids,such as Glu and Asp,and basic amino acids,such as Arg and Lys.In-vivo efficacy of MBP and Fe2+ in improving iron deficiency anemia(IDA) was determined through animal studies.ICR mice were randomly divided into 7 groups:Group 1 was the normal control group,and the other 6 groups were set up by IDA model of iron deficiency diet,which were divided into 1 iron deficiency model group and5 iron deficiency groups.The iron deficiency groups were supplemented with iron glycine(3.5 mg Fe/kg BW) and MBP co-absorption of Fe2+(MBP:FeSO4=8:1,3.5 mg Fe/kg BW),and low,medium and high doses of MBP-Fe(1.16,1.75,3.5 mg Fe/kg BW) respectively once per day via intragastric administration for 28 days.The results showed that the serum iron,hemoglobin and body weight of mice in the high dose MBP-Fe and MBP co-absorption Fe2+ group(3.5 mg Fe/kg BW) were significantly increased(P <0.05),and returned to the normal level,and the spleen coefficient was significantly decreased,compared with the ferrous glycine group.In conclusion,both MPH-Fe and MPH co-absorption of Fe2+ have significant improvement on IDA after reaching the effective amount,and MPH-Fe has the better effect.
