鳄鱼血粉片对环磷酰胺、顺铂致小鼠免疫低下与肾损伤的保护作用研究

【摘要】 目的考察鳄鱼血粉片(CBT)对肿瘤化疗药物环磷酰胺、顺铂造成免疫低下及肾损伤的保护作用。方法分别腹腔注射环磷酰胺(30 mg/Kg)、顺铂(3 mg/Kg)构建小鼠免疫低下、肾损伤模型,灌胃给予CBT低、中、高剂量(0.16、0.33、1.0g/Kg BW)4周后,测定小鼠体重、血浆白细胞(WBC)、红细胞(RBC)、血小板总数(PLT),血红蛋白(HGB)水平,同时测定胸腺及脾脏重量以及肌酐、尿素水平,制备小鼠肾脏切片考察病理改变。结果与环磷酰胺致免疫低下模型组相比,CBT低、中、高剂量能够明显提高小鼠胸腺重量及胸腺指数,提高WBC、RBC及PLT数量,CBT高剂量能够升高小鼠HGB比例,提示CBT对环磷酰胺致小鼠免疫低下具有升高白细胞及红细胞比例,具有较好的提高免疫作用。顺铂致肾损伤模型小鼠血清肌酐、尿素氮水平明显升高,肾脏肾小球毛细血管充血;肾小管上皮细胞可见水肿,细胞核变大变圆,出现个别样本肾小球上皮细胞坏死。与顺铂致肾损伤模型相比,CBT低、中、高剂量能够降低血清肌酐、尿素氮水平,小鼠肾脏肾小管上皮细胞可见轻度水肿,但损伤程度比模型对照组轻,提示CBT对顺铂所致的肾损伤具有保护作用。

【Abstract】 Objective To investigate the protective effects of crocodile blood powder tablet(CBT) on immunosuppression and renal injury caused by tumor chemotherapy drugs cyclophosphamide and cisplatin. Methods The immune depression and renal injury models of mice were established by intraperitoneal injection of cyclophosphamide(30 mg/Kg) and cisplatin(3 mg/Kg). After intragastric administration of CBT at low, medium and high doses(0.16, 0.33, 1.0 g/Kg BW) for 4 weeks, the body weight, total number of white blood cells, red blood cells and platelets in plasma and hemoglobin level were measured. The weight of thymus and spleen and the levels of creatinine and urea were also tested. Mouse kidney sections were prerared to investigate the pathological changes induced by cisplatin.Results Compared with cyclophosphamide induced immunosuppression model, low, medium and high doses of CBT could significantly increase the weight and index of thymus, increase the number of WBC, RBC and PLT, and high doses of CBT could increase the proportion of HGB in mice, suggesting that CBT had a better protective effect on cyclophosphamide induced immunosuppression in mice. In model group mice induced by cisplatin, the levels of serum creatinine and urea increased significantly, renal glomerular capillary congestion,edema of renal tubular epithelial cells, enlargement and roundness of nuclei, and necrosis of glomerular epithelial cells in individual samples were observed. Compared with the model group, low, medium and high doses of CBT can reduce the levels of serum creatinine and urea, mild edema was observed in renal tubular epithelial cells of mice, but the degree was lighter than that of model control group,suggesting that CBT has a protective effect on cisplatin induced renal injury.
