SPG膜乳化法制备量子点荧光编码微球及分析应用

【摘要】 SPG膜乳化法是通过外加氮气压力的作用下将分散相压入膜孔进入连续相而形成乳液。通过对氮气压力、连续相转速、分散相浓度以及不同的量子点对微球的影响,发现了在最优条件下制备的微球粒径均一分散。研究了不同的量子点制备方法以及量子点加量方式对微球均一性影响。在微球表面包裹Fe3O4磁性纳米粒子后制得了磁性微球,其荧光性能大大降低,但其磁性很好与生物活性大幅度提升。将制备的磁性微球通过在外加磁场的定向控制下可以快速且高特异性的将目标生物分子从复杂的生物体系中分离出来,不仅具有磁分离的简便性,同时通过在磁性微球表面修饰特异性抗体使其具备分离的高特异性,拥有双重优势,因此扩宽了微球在生物医学领域的应用。相比较于传统的乳化法如机械搅拌、超声乳化、高速均质乳化等,SPG膜乳化法的操作简单、所制备乳滴粒径均一性好、乳液稳定性好、乳化条件温和、重复性好,有望于推广大规模生产应用。
【Abstract】 The SPG membrane emulsification method is to press the dispersed phase into the membrane pores into the continuous phase by applying nitrogen pressure to form an emulsion.Through the influence of nitrogen pressure,continuous phase rotation speed,dispersed phase concentration and different quantum dots on the microspheres,we found that the particle size of the microspheres prepared under the optimal conditions is uniformly dispersed.The effects of different quantum dot preparation methods and quantum dot addition methods on the uniformity of microspheres are studied.The magnetic microspheres are prepared by wrapping Fe3 O4 magnetic nanoparticles on the surface of the microspheres.The fluorescence performance of the magnetic microspheres is greatly reduced,but the magnetic properties are very good and the biological activity is greatly improved.The prepared magnetic microspheres can quickly and highly specifically separate target biomolecules from complex biological systems under the directional control of an external magnetic field,which not only has the convenience of magnetic separation,but also can be modified on the surface of magnetic microspheres.Specific antibodies make it have high separation specificity and have dual advantages,thus broadening the application of microspheres in the field of biomedicine.Compared with traditional emulsification methods such as mechanical stirring,ultrasonic emulsification,high-speed homogeneous emulsification,etc.,the SPG film emulsification method has simple operation,good uniformity of the prepared emulsion droplets,good emulsion stability,mild emulsification conditions,and good repeatability,is expected to promote mass production applications.
