基于Ni-MOF复合物/AuNPs/CNTs的柔性可拉伸电化学传感器用于实时监测活细胞释放的多巴胺

【摘要】 传统的电化学生物传感器大多利用刚性和不可拉伸电极制作而成,不能实时监测细胞在机械变形状态下释放的生物信号分子。在此,我们基于镍金属有机骨架复合物/Au纳米粒子包覆碳纳米管(Ni-MOF复合物/AuNPs/CNTs)研制了一种可拉伸柔性电化学传感器,用于实时灵敏地检测C6活细胞释放的多巴胺。首先采用一步煅烧的方法,将Ni、NiO和碳框架引入二维Ni-MOF纳米片表面,得到了Ni-MOF复合材料。将Ni-MOF复合物/AuNPs和CNTs分步沉积于PDMS薄膜上,构建的可拉伸电化学传感器具有优良的电化学性能,线性范围为50 nm~15μM,灵敏度为1250 mA·cm2·M-1。此外,可拉伸电极还表现出良好的细胞相容性,C6细胞可以在膜电极上进行培养和增殖,具有较高的活性和较强的粘附性,因而可以实时监测C6活细胞在自然状态和拉伸状态下通过化学诱导释放的DA。

【Abstract】 Existing electrochemical biosensing platforms,using traditional rigid and un-stretchable electrodes cannot real-time monitoring the biological signaling molecules released by cells in a mechanical deformed state.Here,a stretchable and flexible electrochemical sensor was developed based on the nickel metal-organic framework composite/Au nanoparticles coated carbon nanotubes(Ni-MOF composite/AuNPs/CNTs) for sensitive detection of dopamine(DA) released by C6 living cells in real-time.Ni-MOF composite was obtained by introducing Ni,NiO and carbon frame onto the surface of 2 D Ni-MOF nanosheets using an efficient one-step calcination method.The hybrid of Ni-MOF composite/AuNPs/CNTs which deposited on PDMS film endowed the sensor with excellent electrochemical performance with a wide linear range of 50 nM~15 μM and a high sensitivity of 1250 m A·cm-2·M-1.Furthermore,the stretchable electrode also displayed good cellular compatibility while C6 living cells could be cultured and proliferated on it with strong adhesion.Therefore,the DA released by C6 living cells with chemical induction in both natural and stretched states was real-time monitored using our stretchable and flexible electrochemical sensor.
