基于ZrO2/CdS八面体的免标记光电化学生物传感器研究

【摘要】 基于集识别与放大信号为一体的ZrO2/CdS八面体,我们构建了一种免标记、近零背景的光电化学生物传感器实现了蛋白激酶(PKA)活性的灵敏检测,并将其应用于PKA抑制剂的筛选。以Zr-MOF(UiO-66)为前驱体成功合成了尺寸均匀,比表面积大的ZrO2八面体。采用CdS与ZrO2八面体复合后,其光电转换效率显著提升。ZrO2/CdS不仅可以与PKA催化多肽产生的磷酸基团特异性结合,而且可以作为信号探针产生光电信号。该生物传感器对PKA的检测呈现出高灵敏度,检出限约为0.00035 U mL-1 (S/N=3),且在与激酶有关的生命科学和药物领域显示出潜在的应用前景。
【Abstract】 Based on the ZrO2/CdS octahedra that integrate recognition and signal amplification,we constructed a label-free,near-zero background photoelectrochemical biosensor to detect the activity of protein kinase(PKA) and applied it to inhibitors of screening.The porous ZrO2 octahedra with uniform size and large surface area were successfully synthesized by taking Zr-MOF(UiO-66) as the precursor.After the modification of CdS,the photoelectric conversion efficiency of ZrO2 octahedra is significantly improved.The prepared CdS/ZrO2 octahedron can not only specifically interact with phosphorylated peptide,but also can act as signal probe to produce sharp photocurrent.The developed sensor shows high sensitivity for the detection of PKA,with a detection limit of about 0.00035 U mL-1(S/N=3),and shows potential application prospects in the life sciences and pharmaceutical fields related to kinases.
