双波长发射、荧光增强型、特异性检测Cys及其生物成像

【摘要】 本文设计了一种新的双波长"开启"传感器,在检测硫醇的过程中,该探针可以特异性检测内源性和外源性的半胱氨酸。其利用萘基羧基荧光素作为荧光团和丙烯酰基作为半胱氨酸特异性检测的反应位点。在含有该探针的系统中,所加入的半胱氨酸可以诱导明显的双重开启荧光发射,这归因于硫醇和丙烯酰基的亲核反应释放荧光团。双信号荧光响应检测系统使该探针成为半胱氨酸检测的可靠工具,并且该探针的水溶性极好,检出限很低(0.58μM),而且已经成功应用于Hela细胞中的半胱氨酸成像,其促进探针成为理解半胱氨酸相关疾病的病理学的潜在工具。

【Abstract】 In this paper, a new dual-wavelength"turn-on"sensor is designed to specifically detect endogenous and exogenous cysteine during the detection of thiols. It utilizes naphthyl carboxy fluorescein as a fluorophore and an acryloyl group as a reaction site for cysteine-specific detection. In systems containing this probe, the added cysteine can induce significant double turn-on fluorescence emission due to the nucleophilic reaction of the thiol and acryloyl groups to release the fluorophore. The dual-signal fluorescence response detection system makes this probe a reliable tool for cysteine detection, and the probe has excellent water solubility, low detection limit(0.58 μM), and has been successfully applied to the cystine imaging of Hela cells, which promotes the probe is a potential tool for understanding the pathology of cysteine-associated diseases.
