Aptamer功能化DNA四面体纳米诊疗探针用于乳腺癌相关piRNA的成像及治疗

【摘要】 PiRNA(PIWI-interacting RNAs)作为一种新型的癌症候选标志物,与癌症发生与发展的相关分子信号通路有着密切的联系。因此,精准监测piRNA有着重要的意义。与此同时,通过干预piRNA介导的相关癌症信号通路可达到控制和治疗癌症的效果。DNA四面体作为一种毒性低、生物相容性好的DNA纳米结构,在传感器构建和药物递送中实现了广泛应用。本工作以功能化DNA四面体纳米结构为支架,通过结合可与piRNA互补的核酸序列,构建了一种针对piRNA的诊疗一体化纳米探针。通过DNA自组装形成探针支架,再以Aptamer将其功能化使探针更好的靶向癌细胞,探针到达靶细胞内后,pi RNA与四面体探针上可与其结合的序列结合,并将淬灭基团从探针上竞争下来使荧光恢复,实现piRNA成像;与此同时,piRNA与探针结合后,阻碍相应信号通路,抑制肿瘤增殖,引发肿瘤细胞凋亡,对癌症起到一定的治疗效果。
【Abstract】 PIWI-interacting RNAs(piRNAs) have been regarded as novel cancer biomarkers candidate because of their abnormal expression level in various cancer types. DNA tetrahedron is an ideal material for creating various nanoprobe because of its low cytotoxicity and high resistance to cellular permeability. In this study, an aptamer-functionalized DNA tetrahedron has been designed as a theranostic nanoprobe for breast cancer-related piRNA imaging where the target can compete to the quencher oligo for resulting in fluorescence restoration, and molecular pathway-mediated therapy effect on breast cancer.
