基于ExoⅢ辅助靶双重循环和G-四链体的荧光传感用于HIV-DNA检测
Fluorescence biosensor based on Exonuclease Ⅲ-assisted target dual circle and G-quadruplex for HIV-DNA detection

【摘要】 人类免疫缺陷病毒(HIV)是一种逆转录病毒,它会攻击人体的免疫系统,进一步导致获得性免疫缺陷综合症。因此,HIV的分析在临床诊断中非常重要。本实验建立一种基于G-四链体和核酸外切酶III(ExoIII)辅助靶标双重循环信号放大策略快速、简单检测HIV-DNA。在靶标缺失的情况下,G-四链体序列被封闭在发夹中,荧光值低。当添加靶标时,在ExoIII的作用下进行双重循环释放G-四链体序列,NMM嵌入G-四链体中以产生强烈的荧光。该方法通过加靶前后荧光值的变化测定HIV-DNA。在最优的实验条件下,实验结果如下,如图Fig.1. HIV-DNA的检测线性范围为5×10-11?1.5×10-9 mol·L-1,检测限(LOD)为28.4 pmol·L-1(S/N=3)。该方法拥有如下的优点:简单样品处理、操作简便、耗时短等。
【Abstract】 Human immunodeficiency virus (HIV) is a retrovirus,it will attack the body’s immune system,and further leads to acquired immune deficiency syndrome.Thus,the analysis of HIV is extremely important in clinical diagnosis.In this study,we report a new biosensor for the rapidly and simple detection of HIV based on G-quadruplex and Exonuclease III-assisted target to achieve dual circle strategy.In the absence of the target,the G-quadreplex sequences is enclosed in the hairpin,fluorescence value is low.However,when target is added,the dual cycle is carried out and G-quadreplex sequences are released under the action of ExoIII,then the released sequence forms G-quadruplex and NMM is embedded in the G-quadruplex to produce strong fluorescence.HIV is detected by the change of fluorescence value before and after the target.The biosersor has a linear range from 5×10-11 to 1.5×10-9 mol·L-1,the limit of detection (LOD) is 28.4 pmol·L-1(S/N=3).This method has the following advantages:simple sample preparation,convenient operation and short time.
