利用生物素链霉亲和素放大酶联免疫法检测水产品中MG残留
Rapid Detection of Malachite Green in Aquatic Products by BA-ELISA

【摘要】 孔雀石绿(Malachite green,MG)在水产品养殖中常被用作抗菌剂、杀虫剂和防腐剂,该物质容易在生物体内蓄积,并具有致癌性。虽然MG已被列为渔用禁用药,但在水产品中仍频频检出,严重威胁了消费者的身体健康。本实验在制备生物素化抗体的基础上,建立了基于生物素-链霉亲和素放大的酶联免疫分析法,用于水产品中MG的快速分析检测,优化了抗原抗体工作浓度,温育时间,pH值,盐离子强度,封闭液等实验条件。在最佳实验条件下,该方法的线性范围为0.05 ng/mL-20 ng/mL,最低检测限为0.074 ng/mL,回收率为83.5%-115%,CV为4.53%-13.8%,将新方法用于实际阳性样品中MG的分析检测,结果和LC/MS相比具有较好的相关性。该方法灵敏度高,特异性好,准确度高,可以用于水产品中MG的快速分析筛查。
【Abstract】 Malachite green has been banned in aquaculture for many years because of its bioaccumulation and carcinogenicity. However, there is still concern about its illegal use for aqua-culture production. A biotin–streptavidin-amplified enzyme-linked immunosorbent assay(BA–ELISA) was established for MG detection in this study. The experimental conditions such as working concentration of antigen and antibody, incubation time, pH value, salt ion intensity and blocking solution were optimized. The linear range was 0.05 ng/mL-20 ng/mL, the LOD was 0.074 ng/mL, the recovery was 83.5%-115%, and the CV was 4.53%-13.8%. This method has high sensitivity and good specificity, and it can be used for rapid analysis and screening of MG in aquatic products.
