微囊藻毒素的免疫传感器研究
Research on Immunosensor of Microcystins

【摘要】 以微囊藻毒素(MC-LR)为检测研究目标,构筑其紫外可见免疫传感体系[1,2]。MC-LR的适配体和抗体分别功能化PDA@CuNPs(铜纳米粒子包覆聚多巴胺球)和Fe3O4@SiO2(二氧化硅包覆的磁性纳米粒子),进而双位点特异性识别结合MC-LR的相应靶点,形成三明治夹层型复合物;磁性快速分离并将复合体中的铜转化为离子(Cu2+),进一步与双环己酮草酰二腙(BOC)反应,形成turn-on型紫外可见吸收响应。通过TEM、SEM、TGA和FTIR等对Fe3O4@SiO2、PDA@CuNPs复合物进行表征。将实验结果与近十年的MC-LR文献指标相比较,该传感器在检测范围和检测限等方面,都得到了一定的改善,表现出较高的特异性。该检测体系方法简单、灵敏度高、选择性好,实际样品检测性优良。为检测MC-LR提供了潜在的有效方法,具有一定的借鉴意义。
【Abstract】 Microcystins (MC-LR) was selected as a detection target to construct its UV-vis immunosensor.The PDA@CuNPs and Fe3O4@SiO2 were functionalized with the aptamer and antibody of MC-LR respectively,the specific two-site recognition combined the corresponding target of MC-LR and forming a sandwich complex.After rapidly magnetic separation,the copper in the complex was converted into ions (Cu2+),and further reacted with bicyclohexanone oxaloyl dioxime (BOC) to form a turn-on UV-vis signal.The Fe3O4@SiO2 and PDA@CuNPs complexes were characterized by TEM,SEM,TGA and FTIR.Comparing the experimental results with the MC-LR literature of the past decade,the sensor has been improved in terms of detection range and detection limit,showing a high specificity.The detection system has the advantages of simple method,high sensitivity,good selectivity and sample detection performance.For the detection of MC-LR,it provided a potentially effective method.
