自组装纳米颗粒用于中和蜂毒肽毒性的研究

【摘要】 抗体是用于特异性分子识别的常用工具,但其不易生产、成本高、稳定性差、易变性等限制了其应用范围,因此设计新的抗体替代物具有重要意义。聚合物纳米颗粒具有成本低、易制备、稳定等优点,设计与特定生物大分子结合的聚合物纳米颗粒作为抗体的替代物具有重要的应用价值。在本工作中,基于β-环糊精聚合物与金刚烷衍生物的主客体识别作用,我们设计了一个包含82种自组装超分子纳米颗粒(SSNP)的人工抗体文库,通过溶血实验筛选能中和蜂毒肽溶血毒性的SSNP。实验结果表明,我们获得了8种能中和蜂毒肽溶血毒性的SSNP,中和效率均在80%以上。这种筛选人工抗体的方法为开发新的解毒剂提供了一种全新的视角。
【Abstract】 Antibodies are common tools for specific molecular recognition. However, they are high cost, poor stability and easy denaturation. Thus, generating new alternatives for antibodies is significance. Since polymer nanoparticles have the advantages of low cost, easy preparation and stability, it is of great relevance to applications to design polymer nanoparticles capable of binding to specific biomacromolecules as artificial antibodies. In this work, we designed an artificial antibodies library containing 82 self-assembled supramolecular nanoparticles(SSNPs) based on the host–guest inclusion of β-cyclodextrin polymer and adamantane derivatives, and selected SSNPs which were capable of suppressing the toxicity of melittin by hemolytic activity neutralization assay. The results showed that there were 8 SSNPs with neutralization efficiency higher than 80%. This method of screening artificial antibodies provides a new perspective for the development of new antidotes.
