中国蜂胶改善STZ诱导糖尿病大鼠的早期糖尿病视网膜病

【摘要】 蜂胶具有降低血糖和抑制糖尿病肾病病变的作用,但其是否有改善糖尿病视网膜病的作用还未可知。我们探究了在STZ诱导的糖尿病大鼠中,中国蜂胶对早期糖尿病视网膜病的效果。用乙醇提取的中国蜂胶饲喂糖尿病大鼠12周后,进行空腹血糖、糖化血红蛋白、丙二醛、活性氧簇和活性氮簇浓度的测量,以及视网膜组织切片的苏木素伊红染色、免疫荧光染色观察和透射电子显微镜观察。结果显示,中国蜂胶能够降低空腹血糖和糖化血红蛋白的浓度,以及丙二醛、活性氧簇和活性氮簇的浓度。此外,中国蜂胶抑制了感光细胞损伤和视网膜增厚,并且通过增加紧密连接蛋白occludin和ZO-1的表达抑制血视网膜屏障的渗漏。表明中国蜂胶能够通过保护血视网膜屏障的途径改善糖尿病大鼠的早期糖尿病视网膜病。

【Abstract】 Propolis has been shown to reduce the level of blood glucose and suppress the histopathological changes in diabetics.However,it still remains unknown if propolis has similar effect on diabetic retinopathy(DR).Our study was aimed to evaluate the effect of Chinese propolis(CP) on early DR in streptozotocin(STZ)-induced diabetic rats.Ethanol extract of CP was given to diabetic rats by oral intubation for 12 weeks.The concentrations of fasting blood glucose(FBG),glycated hemoglobin(HbAlc),malondialdehyde(MDA),reactive oxygen species(ROS) and reactive nitrogen species(RNS) were measured.Pathological examinations including hematoxylin and eosin(HE) staining,transmission electron microscopy(TEM) and immunofluorescence were also conducted to provide further evidence of CP’ s effect on early DR.Results:CP was able to attenuate diabetes via directly decreasing the levels of FBG and HbAlc,which also resulted in the reduction of MDA,ROS and RNS.Furthermore,CP could protect against the damages of photoreceptor cells as well as the retinal thickening.And the inhibition of blood-retinal barrier(BRB) leakage was also observed in CP treated diabetic rats along with the inhibition the loss of tight junction proteins(occludin,ZO-1).These results suggested that CP has an ameliorating effect on early DR by inhibition of blood-retinal barrier breakdown.
