液相色谱-二级质谱联用法定量测定蜂王浆主要脂肪酸的研究

【摘要】 为了建立蜂王浆脂肪酸定量分析的方法,本研究测定了10-羟基-2-烯葵酸(10-hydroxy-2-decenoic acid,10-HDA)等8种蜂王浆主要脂肪酸含量。首先,通过生物化学分级分离方法从蜂王浆冻干粉中制备了蜂王浆脂质组;其次,通过阴离子交换色谱方法纯化了蜂王浆脂肪酸提取物;最后,通过液相色谱-二级质谱联用(LC-MS/MS)方法定量测定了蜂王浆脂肪酸提取物的10-HDA等8种蜂王浆主要脂肪酸含量。结果显示,基于多反应监测数据的10-HDA等8种蜂王浆主要脂肪酸定量值百分比(%)分别为1.5678、0.3408、0.1097、0.0268、0.1321、0.0381、0.0739以及0.0917。这表明,LC-MS/MS可以成为定量测定蜂王浆主要脂肪酸的检测方法。

【Abstract】 To set up a quantitative method for determination of main fatty acids in royal jelly,we measured eight main fatty acids such as 10-hydroxy-2-decenoic acid(10-HDA) extracted from royal jelly.First of all,lipidome was extracted from lyophilized royal jelly by biological fractionation method.Furthermore,main fatty acids were purified from the extracts by anion chromatography method.Finally,eight main fatty acids such as10-HDA were quantitatively measured by LC-MS/MS.It is shown from the study that measured values of these fatty acids were characterized as percentage by multiple reaction monitoring dataset,including 1.5678,0.3408,0.1097,0.0268,0.1321,0.0381,0.0739 and 0.0917.It is suggested from these results that LC-MS/MS could be an alternative method for quantitative determination of main fatty acids in royal jelly.
