黑木耳黑色素提取及体外抗氧化活性初步研究

【摘要】 以东北优质"黑29"黑木耳为原料,以黑色素粗提物收率为评价指标,在单因素试验基础上,通过响应面法研究确定黑木耳黑色素提取最佳工艺条件,并对其进行体外抗氧化活性研究。结果:黑木耳黑色素提取最佳工艺条件为浸提体系pH值13.0,温度69℃,浸提时间59min,黑色素粗提物收率为2.877%,纯度为88.2%,经实验确定其体外抗氧化活性较强,黑木耳黑色素浓度0.24mg/mL对DPPH自由基的清除率大于50%;浓度为0.375mg/mL时,对超氧阴离子自由基清除率达39.79%;浓度为12~60μg/mL时,黑木耳黑色素对羟基自由基的清除率低于10%。

【Abstract】 Taking the northeast high-quality Auricularia auricula(Heiwei 29) as experimental material, extract yield of melaninas the evaluation index, on the basis of single factor experiment, by response surface method, the optimum conditions for the extraction of melanin from A. auricula were determined,its antioxidant activities werealso studied. Results: It found out that the optimum conditions for the extraction of melanin from A. auricula were water bath extraction pH13.0, the temperature of 69℃, extracting time of 59 minutes, 2.877% extract yield of melanin, and purify was 88.2%. The experiment also showed that antioxidant activities in-vitro were strong, the rate of scavenging free radical DPPH was more than 50% when A. auricula melanin concentration was 0.24 mg/mL, the rate of scavenging free radical superoxide anion reached up to 39.79% when its concentration was 0.375 mg/mL; the rate of scavenging free radical hydroxyl was less than 10% when its concentration was in range 12~60 μg/mL.
