基于网络药理学探讨柴胡-黄芩药对治疗功能性消化不良的作用机制

【摘要】 通过临床调研,统计出柴胡-黄芩为治疗功能性消化不良最常用药对,在临床结果的基础上,利用网络药理学和文献调研,按照"药物-靶点-疾病"关联性研究,分析功能性消化不良相关蛋白的互作网络,构建药对活性成分-预测靶点网络,进而对功能性消化不良相关的柴胡-黄芩药对活性成分-潜在靶点网络进行分析。分析出主要的潜在靶点网络包含18个靶点,关键靶点涉及乙酰胆碱酯酶、细胞色素P450、5-羟色胺受体2B、一氧化氮合酶2、b细胞CLL/淋巴瘤2、丝裂原活化蛋白激酶8、溶质载体家族6成员3等。对潜在靶点进行基因本体功能富集分析得出主要的生物过程及分子功能有调节蛋白质定位的建立、调节蛋白质定位、单一生物代谢过程、对化学品的反应、对含氧化合物的反应及酶结合等。进行KEGG富集通路分析主要通路有内分泌抵抗,癌症的途径,GnRH信号通路,5-羟色胺能突触,钙信号通路,CAMP信号通路等。结果表明柴胡-黄芩药对可以通过多成份、多靶点、多途径共同发挥治疗功能性消化不良的作用。

【Abstract】 Through clinical investigation,Bupleurum-Huangqin is the most commonly used drug pair for treating functional dyspepsia.Based on the clinical results,network pharmacology and literature research were used to analyze the interaction network of proteins associated with functional dyspepsia according to the "drug-target-disease" relevance research,and to build an active component-predicted target network so as to analyze the active component associated with functional dyspepsia in Bupleurum-Huangqi-potential target network.It is analyzed that 18 targets were contained in the main potential target network.The key targets involved Acetyl cholinesterase,Cytochrome P450,family,subfamily D,polypeptide 6,5-Hydroxytrptamine Receptor 2 B,Nitric oxide synthase 2,B-cell CLL/lymphoma 2,Mitogen-Activated Protein Kinase 8,Solute Carrier Family 6 Member 3,etc.Gene ontology functional enrichment analysis of potential targets revealed that the main biological processes and molecular functions include regulation of establishment of protein localization,regulation of protein localization,single-organism metabolic processe,reactions to chemicals,reactions to oxygen-containing compound,enzyme binding and so on.According to KEGG enrichment pathway analysis,the main pathway include endocrine resistance,pathway in cancer,GnRH signaling pathway,serotoninergic synapse,calcium signaling pathway,and cAMP signaling pathway.The results showed that Bupleurum-Huangqin can play a role in the treatment of functional dyspepsia through multiple components,multiple targets and multiple pathways.
