符合中医证候的血虚便秘小鼠模型的建立与评价

【摘要】 目的:建立符合中医证候的血虚便秘小鼠模型。方法:64只SPF级KM小鼠,随机分为正常组、血虚组、便秘组和血虚便秘组,每组10只。血虚组小鼠皮下注射乙酰苯肼60 mg/kg+腹腔注射环磷酰胺40 mg/kg,便秘组小鼠灌胃复方地芬诺酯25mg/kg,血虚便秘组小鼠皮下注射乙酰苯肼60 mg/kg+腹腔注射环磷酰胺40 mg/kg+灌胃复方地芬诺酯25 mg/kg,正常组小鼠皮下注射+腹腔注射+灌胃等量生理盐水,造模连续28 d。观察各组小鼠血虚和便秘证候,检测体重、血常规、首粒黑便排出时间(TFBF)、粪便含水量(FWC)和结肠含水量(CWC);取各组小鼠结肠组织进行HE和AB-PAS染色。结果:与正常组比较,血虚组小鼠随造模时间延长,体重增长缓慢,外周血WBC、RBC、HGB和HCT均显著降低(P<0.05,P<0.01),粪便质软,TFBF、FWC和CWC均无显著性变化;便秘组小鼠体重增长稍慢,外周血WBC、RBC、HGB和HCT无显著变化;血虚便秘组体重增长缓慢,外周血WBC、RBC、HGB和HCT均显著降低(P<0.05,P<0.01);便秘组和血虚便秘组小鼠粪便质硬,TFBF显著延长、FWC和CWC均显著下降(P<0.05,P<0.01);血虚小鼠结肠组织形态和黏液分泌与正常组相似,便秘和血虚便秘小鼠结肠粘膜和大肠腺萎缩,黏液分泌减少。结论:乙酰苯肼、环磷酰胺联合复方地芬诺酯能建立符合中医证候的血虚便秘小鼠模型,造模方法可控、简便、易行。

【Abstract】 Objective:To establish and evaluate Xue-Xu with constipation(XC) model in mice conforms to TCM symptoms and signs.Methods:64 SPF KM mice were randomly divided into four groups:normal group,Xue-Xu group,Constipation group and XC group.The mice in the Xue-Xu group were subcutaneous injected(sc) Acetylphenyhydrazine(APH)and intraperitoneal injected(ip) cyclophosphamide(CPA),the mice in the Constipation group were irrigated(ig) Diphenoxylate,the mice in the XC group were administrated APH and CPA and Diphenoxylate.the blood deficiency and constipation syndrome,body weights,feces properties,complete blood count,the time of the first black feces(TFBF),the feces water content(FWC) and the colon water content(CWC) were observed.HE and AB-PAS staining were performed.Results:Compared to normal group,the mice in Xue-Xu group:Weight-increasing speed were reduced,the levels of WBC,RBC,HGB,HCT were all decreased,the feces were soft,there were no significant change in TFBF,FWC and CWC;the mice in constipation group:Weight-increasing speed and the levels of WBC,RBC,HGB,HCT were all normally;the mice in XC group:Weight-increasing speed were reduced,the levels of WBC,RBC,HGB,HCT were all decreased;the mice in constipation and XC group:the feces were hard,the TFBF were significantly increased,FWC and CWC were significantly decreased.The morphology and mucus secretion of colonic tissues in Xue-Xu group were close to normal control group,the colonic mucous membranes were narrowed in some extent and the mucus excretion were decreased in Constipation and XC group.Conclusion:It’s a simple and controllable method to establish XC mice model by administrating of APH,CPA and Diphenoxylate.
