植物转基因成分目标序列测序方法研究
Research on target sequencing method for ingredients of genetically modified plants

【摘要】 现有的转基因检测技术多数以PCR扩增为基础,一次只能检测一种外源基因成份,也不能检测未知成分,难以避免转基因检测的假阴性结论。同时,PCR扩增无法区分微生物污染与气溶胶污染,因此,也难以避免转基因检测的假阴性结论。该研究建立了植物转基因成分目标序列测序方法。结果表明,利用441个杂交探针,实现了所有已知的28个转基因元件、73种已知的转基因品系和其它未知转基因品系的检测;55个含有1%的不同外源基因的转基因标准样品均通过本标准的方法检出含有至少一种外源基因成分,成功率100%;对116个转基因阳性样品进行了检测,其中有112个样品被判定含有一种或多种转基因成分,正确率为96.56%。对100个已知的非转基因植物品种的样品进行了测试,均没有发现任何转基因成分,结论的正确率为100%。
【Abstract】 Most of the existing transgenic detection technologies are based on PCR amplification.However,these methods can only detect one exogenous gene component at a time and cannot be used to detect unknown components,so it is difficult to avoid the false negative conclusion of transgenic detection.Meanwhile,PCR amplification could not distinguish microbial pollution from aerosol pollution,which may also result in a false negative conclusion.Therefore,target sequencing method for ingredients of genetically modified plants was established in this research.The results showed that the target sequence of 28 known transgenic elements,73 known transgenic strains and other unknown transgenic strains were detected by using 441 hybrid probes.All the 55 transgenic standard samples containing 1% different exogenous genes were detected to contain at least one exogenous gene by this standard method,and the accuracy rate reached for 100%.112 samples of 116 positive transgenic samples tested were confirmed to contain one or more transgenic ingredients with the accuracy rate of 96.56%.No transgenic components were found in 100 known non-transgenic plants with the accuracy rate of 100%.
