【摘要】 目的:采用高效液相色谱法测定人血浆中万古霉素浓度,考察不同沉淀剂对万古霉素提取率的影响。方法:色谱柱:Wondasil^?C₁₈-WR （4.6×150mm,5μm）,流速:1.0mL·min^-1,检测波长:236nm,内标:去甲万古霉素,分别考察流动相为甲醇-磷酸二氢钾缓冲液（50mM,pH 3.2）与乙腈-磷酸二氢钾缓冲液（50mM,pH 3.2）时,不同沉淀剂（10%高氯酸80μL,10%高氯酸80μL-二氯甲烷200μL,10%三氯醋酸100μL,乙腈-6%高氯酸（1:6） 200μL）对血浆样品提取率的影响。结果:结果显示乙腈-6%高氯酸（1:6）作为沉淀剂对万古霉素提取率最好。血浆万古霉素浓度在5～100μg·mL^-1内线性良好,Y=0.0210X-0.0007（r=0.9999,n=6）,日内RSD<2.79%,日间RSD<5.17%,绝对回收率为79.91～91.71%。结论:乙腈-6%高氯酸（1:6）作为沉淀剂,提取率高、重现性好、操作简便、灵敏,适用于万古霉素血药浓度测定。更多还原

【Abstract】 Objective:To establish a method for determination of vancomycin in human plasma by HPLC,and to investigate the effects of different precipitating agent to vancomycin extraction rate.Methods:Separation was carried out on a Wondasil^? C₁₈-WR column（4.6 x150 mm,5μm）.flow rate was 1.0 mL·min^-1,and UV detector wave length was 236 nm.Norvancomycin was used as the internal standard.The mobile phase were investigated methanol-phosphate buffer（50 mM,pH 3.2） and acetonitrile-phosphate buffer（50 mM,pH3.2）.Different precipitation agents（10% perchloric acid 80μL,10% high acid 80μL-dichloromethane 200μL,10% TCA 100μL,acetonitrile-6% perchloric acid（1:6） 200μL） were used to investigate the affect of plasma sample extraction rate.Results:The results showed that acetonitrile-6% perchloric acid（1:6） as a precipitating agent,the extraction rate was best.The linear range of vancomycin was 5-100μg·mL^-1 with a linear equation Y=0.0210 X-0.0007（r=0.9999,n=6）.The intra-day and inter-day precisions were less than 2.79% and 5.17%,respectively.The absolute recovery was 79.91～91.71%.Conclusion:The acetonitrile-6% perchloric acid（1:6）as a precipitating agent was high extraction rate,reproducible,simple and sensitive,which can be used in the determination of plasma concentration.更多还原
