响应面法优化酶解南极磷虾制备亚铁螯合肽的工艺研究
Optimization of enzymatic preparation of iron-chelating peptides from Antarctic krill(Euphausia superba) by response surface methodology

【摘要】 目的:以南极磷虾蛋白为原料,运用响应面法酶解制备具有较高亚铁螯合活性的南极磷虾酶解物。方法:以亚铁螯合率为主要指标,水解度为辅助指标,筛选最佳蛋白酶,并通过单因素试验考察酶解温度、酶解时间、加酶量和pH值对酶解物亚铁螯合能力的影响。在单因素试验基础上,利用响应面法进行酶解工艺条件的优化。结果:Alcalase 2.4L是酶解南极磷虾蛋白的最适用酶,最佳酶解工艺条件为酶解温度57.6℃,加酶量5.5%(质量分数),pH值8.8,酶解时间300 min。在此条件下,酶解物的亚铁螯合率预测值为78.88%,验证值为77.76%,实测结果与预测值符合良好。此外,氨基酸分析表明具有较高亚铁螯合活性的酶解物中谷氨酸、赖氨酸、亮氨酸、精氨酸及天冬氨酸含量较高。讨论:在最佳酶解条件下得到的南极磷虾酶解物中几种特定氨基酸含量占总氨基酸的比例较大,这几种氨基酸中的特定基团与残基可能是主要的亚铁螯合位点。
【Abstract】 Objective:Antarctic krill protein was used as raw material to prepare iron-chelating peptides by response surface methodology(RSM).Methods:The optimal protease was selected on the iron-chelating rate and the hydrolysis degree of hydrolysates.The effects of enzymolysis temperature,enzymolysis time,enzyme dosage and pH were evaluated by single factor experiment.The RSM was applied to optimize the enzymatic hydrolysis process based on the results of single factor experiments.Results:Alcalase 2.4 L showed the highest hydrolysis degree and iron-chelating rate.The optimal enzymolysis conditions were enzymolysis temperature of 57.6 ℃,enzyme dosage of 5.5%(W/W),pH 8.8 and enzymolysis time of 300 min.On this condition,the predicted iron-chelating rate was 78.88%,which was in good accordance with the actual average value of 77.76%.In addition,the amino acid composition analysis indicated that the obtained hydrolysate possessed high contents of glutamic acid,lysine,leucine,arginine and aspartic acid.Discussion:The contents of several specific amino acids were abundant in hydrolysate prepared from Antarctic krill under the optimal enzymatic condition.The specific groups and residues of these certain amino acids were probably able to participate in the combination with ferrous ions.
