黑果枸杞花色苷的纯化及对人体肠道菌群的调节作用

【摘要】 黑果枸杞是近年来新发掘的一种重要的野生植物资源,具有抗氧化,抗炎症,降血脂和抗肥胖等功效,而花色苷是黑果枸杞中主要的活性成分之一。我们利用高速逆流色谱(HSCCC)分离纯化得到了黑果枸杞中的主要花色苷:矮牵牛素3-O-芸香苷-5-O-葡萄糖苷(P3G),并通过体外模拟发酵研究了P3G对人体肠道菌群的影响。结果表明,乙腈/正丁醇/甲基叔丁基醚/水/三氟乙酸(1:2:2:5:0.01)作为HSCCC的溶剂体系有利于黑果枸杞中P3G的分离纯化,纯度达到了96%。P3G在体外模拟发酵过程中不仅能提高双歧杆菌和乳酸杆菌的丰度,降低Firmicutes/Bacteroidetes值(从116.01±16.27降低到67.16±11.44),还能促进短链脂肪酸的生成,从(21.76±2.10)mmol/L增加到(51.61±4.31)mmol/L。因此,P3G对人体肠道菌群的调节有着积极的作用。

【Abstract】 Lycium ruthenicum Murray is an important wild plant resource newly discovered in recent years.It has many health benefits,such as antioxidant,anti-inflammatory,hypolipidemic and anti-obesity effects.Anthocyanins are the main active components in Lycium ruthenicum Murray.We purified the main anthocyanin(petunidin-3-O-[rhamnopyranosyl-(trans-P-coumaroyl)]-5-O-[β-D-glucopyranoside,P3G) in Lycium ruthenicum Murray by high-speed countercurrent chromatography(HSCCC).The effects of P3G on human intestinal microbiota were also studied by simulated fermentation in vitro.The results showed that acetonitrile/n-butanol/tert-butyl methyl ether/water/trifluoroacetic acid(1:2:2:5:0.01) as the solvent system of HSCCC was the most suitable for the purification of P3G from Lycium ruthenicum Murray,and the purity reached 96%.In the simulated fermentation in vitro,P3G not only increased the relative abundance of Bifidobacterium and Lactobacillus,but also decreased the Firmicutes/Bacteroidetes ratio(from 116.01±16.27 to 67.16±11.44) and promoted the production of short-chain fatty acids,from(21.76±2.10) mmol/L to(51.61±4.31) mmol/L.Therefore,P3G has a positive effect on the regulation of human intestinal microbiota.
