燕麦β-葡聚糖对大豆油消化行为及脂肪酸转运特性的影响

【摘要】 目的:基于体外胃肠消化模型和Caco-2细胞单层上皮模型,研究不同剂量燕麦β-葡聚糖对大豆油消化及吸收特性的影响。方法:通过pH-state法、激光纳米粒度仪、GC-MS等方法对大豆油体外胃肠消化产物的脂肪酸释放率、粒径、胰脂肪酶活性及不同游离脂肪酸的比例进行测定;基于Caco-2细胞模型采用透射电镜负染法、RT-PCR分析大豆油体外消化产物对脂肪酸转运特性的影响。结果:胃肠消化中,燕麦β-葡聚糖添加量由0%增加到0.2%时,消化产物中游离脂肪酸从30.92%降低到18.6%;消化产物的平均粒径显著增大,肠消化中脂肪酶的活性呈下降趋势;消化产物中不饱和脂肪酸所占的比例增大;添加燕麦β-葡聚糖的消化产物作用于Caco-2细胞后,培养液中CM数量降低,且粒径增大,细胞内脂肪酸吸收及转运蛋白FATP4,DGAT1,MTP和L-FABP mRNA的表达量均显著低于对照组(P<0.05)。结论:燕麦β-葡聚糖通过改变大豆油的消化特性,使Caco-2细胞单层中脂肪酸吸收及转运蛋白表达降低,改变了脂肪酸的吸收过程,降低了细胞对脂质的吸收和转运。

【Abstract】 Objective:Based on a simulated gastrointestinal tract(GIT) model and Caco-2 cells monolayer model,we studied the impact of oat B-glucan on soybean oil digestion and absorption characteristics in the digestion system.Methods:By using pH-state method,laser particle size analyzer and GC-MS,the release rate,particle size,pancreatic lipase activity and the proportion of different free fatty acids of soybean oil gastrointestinal digestive products were determined by the methods.Based on Caco-2 cell model,TEM negative staining and RT-PCR were used to analyze the effect of soybean oil in vitro digestion products on fatty acid transport properties.Results:In gastrointestinal digestion,when the addition amount of oat β-glucan was increased from 0.0% to 0.2%,the percent of free fatty acids released in digestion products decreased from 30.92% to 18.60%.The average particle size of the stomach,intestinal digestion products increased significantly.Lipase activity in the intestinal digestion system showed a downward trend.The proportion of unsaturated fatty acids of soybean oil digestion product increased.After the soybean oil digestion product in digestion system with adding oat β-glucan to Caco-2 cells,the number of chylomicrons(CM) observed by TEM was more and the particle size was larger than the control;meanwhile,the expression level of fatty acid absorption and transport related proteins FATP4,DGAT1,MTP and L-FABP mRNA was reduced significantly(P < 0.05).Conclusion:Oat-glucan altered the digestive properties of soybean oil.These effects caused fatty acid absorption in the Caco-2 cell monolayer and transport-related protein expression were significantly reduced,thereby changing the absorption process of fatty acids,reducing cell lipid absorption and transport rate.
