血小板萃取结合HPLC分析筛选中药延胡索的抗血小板活性生物碱

【摘要】 目的筛选活血化瘀中药延胡索中的抗血小板聚集生物碱。方法采用血小板萃取方法富集延胡索中能与血小板特异性结合的成分,结合HPLC-MS分析手段对筛选出的成分进行鉴别,并进行体外抗血小板活性验证。结果共筛选出延胡索生物碱中能与血小板特异性结合的5个化合物,分别是海罂粟碱、脱氢紫堇碱、四氢小檗碱、四氢黄连碱和延胡索甲素,都具有较强的体外抗血小板聚集作用,IC50值分别为49.057、34.914、33.547、84.261和54.164μg mL-1。结论血小板萃取结合HPLC分析是快速有效的中药活性成分筛选方法。
【Abstract】 OBJECTIVE To screen potential anti-platelet aggregation compounds in the total alkaloids extract(TAE) of Rhizoma Corydalis. METHODS Employing platelet bio-specific extraction to enrich compounds which can be specifically bond to platelets in TAE, the HPLC analysis was used for constituent identification and quantification, further anti-platelet aggregation activity of the screened compounds were confirmed in vitro by assay. RESULTS Five alkaloids, which were identified as glaucine, dehydrocorydaline, canadine, tetrahydrocoptisine and corydaline by their MS spectra, can be specifically extracted by platelets. And their IC50 values for platelet aggregation inhibition were 49.057, 34.914, 33.547, 84.261 and 54.164 μg mL-1, respectively. CONCLUSION It is suggested that bio-specific extraction combined with LC-MS analysis is a useful tool for screening of active compounds from Chinese medicines.
