猪萨佩罗病毒感染对断奶仔猪小肠粘膜形态及抗病毒免疫功能的影响

【摘要】 为探讨猪萨佩罗病毒(PSV)感染对断奶仔猪小肠粘膜形态及抗病毒免疫功能的影响。9头21日龄的断奶大白仔猪随机分成3组,分别口服低感染剂量PSV毒株、高感染剂量PSV毒株和DMEM,感染3天后处死,剖检并观察小肠病理学变化;组织病理学观察仔猪小肠粘膜形态的变化;免疫组织化学、组织化学对肠道组织内的免疫细胞定位及数量分析;荧光定量PCR检测肠道模式识别受体通路相关因子的变化。PSV感染后的仔猪临床症状仅表现为轻微腹泻;组织病理学检查结果显示PSV感染导致肠绒毛断裂脱落,固有层炎性细胞浸润,派尔结淋巴细胞减少,黏膜下层水肿,肌层轻微断裂;小肠各肠段CD3+T淋巴细胞、CD4+T淋巴细胞、杯状细胞、肥大细胞数量均有不同程度的减少,各肠段CD8+T淋巴细胞数量均有不同程度的增多,回肠中B淋巴细胞数量减少,巨噬细胞数量增多;小肠中TLR4、RIG-1、MVA-5以及IFN-β的m RNA表达水平显著性升高。结果表明,PSV感染后在小肠绒毛上皮细胞和淋巴细胞定植,改变了猪肠道免疫细胞的数量、位置和形态,激活了肠道PRR通路,诱导肠道产生高水平的干扰素,从而激活机体的抗病毒免疫功能。
【Abstract】 To investigate the impact of Porcine Sapelovirus(PSV) infection on the mucosal morphology and antiviral immunity in weaned pig small intestine. Nine 21-day-old weaned large-white piglets were randomly divided into 3 groups. They were given oral administration of PSV strain with low infection dose, PSV strain with high infection dose and DMEM. Pigs in three groups were euthanized three days after inoculation and observed the pathological changes of small intestine; histopathological changes were observed in the pig small intestine; immunohistochemical and histochemical were uesd to analyse the chance of localization and quantitative of immune cells in intestinal tissue; q PCR was used to detect changes in patterns recognition receptor pathway related factors in intestinal. Clinical symptoms indicated that piglets only showed mild diarrhea after PSV infection. Histopathological observations were characterized by intestinal villus rupture, lamina propria inflammatory infiltration, Peyer’ s Patches lymphopenia, submucosal edema, and muscle layer rupture, which were caused by PSV infection. After PSV infection in pigs, the number of CD3+ T lymphocytes, CD4+ T lymphocytes, goblet cells and mast cells in each intestinal segment of the small intestine decreased to varying degrees, and the number of CD8+ T lymphocytes in each intestinal segment increased to varying degrees. The number of B lymphocytes in the duodenum and jejunum increased, while decreased in the ileum. And the number of macrophages increased. PSV infection also significantly enhanced the m RNA expression levels of TLR4, RIG-1,MDA-5 and IFN-β. The results showed that colonization of intestinal epithelial cells and lymphocytes after PSV infection changed the number, location and morphology of intestinal immune cells, activated the intestinal PRR pathway, and induced high levels of interferon in the intestinal tract, so that the body was in active antiviral state.
