红色物种海丰沙塘鳢地理分布及其线粒体基因组研究

【摘要】 海丰沙塘鳢已知地理分布仅限于广东省龙津河和东江流域。本研究在粤东榕江的南河和北河分支分别采集到了数尾沙塘鳢个体,经形态学比对研究,确认为海丰沙塘鳢。同时,通过PCR测序首次获得了海丰沙塘鳢线粒体基因组全序列。海丰沙塘鳢线粒体基因组全长17016bp,包含13个编码基因,22个t RNA,2个r RNA和1个D-loop区。海丰沙塘鳢线粒体基因组与已知沙塘鳢属物种结构相似,也发现了沙塘鳢属特有的SLH基因簇tRNA基因重组结构。比对分析表明,该重组结构是由大规模基因片段复制后部分基因随机丢失导致,该t RNA基因重组结构可以作为沙塘鳢属个体鉴别的特异性分子标记。本研究扩大了红色易危物种海丰沙塘鳢的地理分布记录,并首次获得了其线粒体基因组全序列,为该物种的生物学研究和保护奠定了坚实基础。
【Abstract】 Odontobutis haifengensis Chen, 1985, is restrictly distributed in two rivers in Southern Guangdong Province, China. Two specimens of O. haifengensis were caught from different sites of Rongjiang River. This work provides a partial description and a few morphological measurements of that specimen. These records extend the known distribution range of O. haifengensis eastward in Guangdong Province, China. Meanwhile, the complete mitochondrial genome of O. haifengensis was sequenced for the first time,which was 17,016 bp in length and included 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs, and a control region. The genome organization was similar to other Odontobutis species. Furthermore, a t RNA gene rearrangement within the SLH cluster was found to be identical to other Odontobutis species, which might result from a tandem duplication and random loss of large-scale gene regions. This study provides useful information that aids in a better understanding of mitogenomic diversity and evolutionary patterns of Odontobutidae species.
