坛紫菜叶状体长度性状的BSR分析及相关SNP位点开发
BSR analysis for length trait of Pyropia haitanensis and development the SNP markers

【摘要】 坛紫菜(Pyropia haitanensis)的经济性状如产量、长度、品质等都属于多基因控制的数量性状,易受环境条件影响,采用常规育种手段存在育种周期长、效率低,而且性状不稳定等诸多问题。开发与目标性状相关的分子标记,进行分子标记辅助育种可以显著加快育种进程,但目前尚未有与坛紫菜经济性状相关的分子标记报道。本研究以坛紫菜DH作图群体中叶状体长度性状分别表现为最长和最短的8个品系为实验材料,构建两个极端混合池,利用BSR技术对混池样品进行转录组测序分析。数据结果显示,长组和短组的差异基因主要富集在物质合成、能量代谢、细胞骨架以及ABC转运蛋白。同时,在80个关联到差异表达基因上的SNP中,有22个SNP可能与坛紫菜长度性状相关。进一步验证发现,通过长短性状相关联的SNP定位到3个有注释的候选基因:α微管蛋白、CBF-B/NF-YA转录因子和假设蛋白。本研究为筛选用于坛紫菜遗传选育和品种改良的高质量SNP标记奠定了基础。
【Abstract】 Several key economic traits(such as yield, thallus length and quality) of Pyropia haitanensis are quantitative traits and usually controlled by multiple genes, and are generally sensitive to changes on environmental conditions. The disadvantage of conventional breeding methods includes long breeding cycle, low efficiency and unstable traits, etc. Through development of target traits related molecular markers, molecular marker-assisted breeding can significantly accelerate the breeding process. However, to date, there is no report on molecular marker which related to the economic traits of P. haitanenis. In the present study, two RNA pools were established based on eight longest and eight shortest strains in the DH population of P. haitanenis. BSR method was performed to analyze the sequences of the RNA pools. The results showed that the differentially expressed genes between long and short groups were mainly enriched in substance synthesis, energy metabolism, cytoskeleton and ABC transporters. This study found 80 SNPs which related to differentially expressed genes, and 22 SNPs were identified to be related to the length traits of P. haitanenis. Further studies revealed three annotated candidate genes: alpha tubulin, CBF-B/NF-YA transcription factor and hypothetical protein. This study laid the foundation for screening high-quality SNP markers for genetic breeding and variety improvement of P. haitanenis.
