坛紫菜不同发育时期的转录组学比较分析

【摘要】 坛紫菜(Pyropia haitanensis)是我国南方沿海传统栽培的一种大型经济海藻。开展坛紫菜生活史的研究是紫菜人工栽培、遗传育种研究的基础,但目前仍缺乏对坛紫菜整个生长发育过程的系统性分析。本实验以坛紫菜野生型品系"北岚岭"的不同发育时期样本为实验材料,通过比较转录组分析初步揭示了坛紫菜生长发育过程中存在的调控机理。数据分析结果显示,丝状藻丝形成双分孢子的过程中,光合作用通过积累有机物用于双分孢子的分化与壳孢子的释放;双分孢子释放壳孢子到壳孢子形成叶状体的过程中,错配修复相关基因参与了壳孢子的减数分裂过程;脯氨酸、肌动蛋白和泛素结合酶等可以提高壳孢子对水体逆境环境的适应性。叶状体细胞分化成生殖细胞再形成合子的过程中,光合作用、基础代谢水平降低,DNA复制和染色质重塑过程相关代谢增强,蛋白质周转加快。果孢子在萌发形成丝状藻丝的过程中,细胞骨架、大分子HSP和植物激素不仅促进新的营养藻丝的形成,还提高了藻体对于水体环境的适应性。此外,磷脂酰肌醇信号系统是坛紫菜丝状体适应不同生长环境的重要信号转导系统。本研究为后续坛紫菜生活史的研究和新品种的选育奠定了理论基础。
【Abstract】 Pyropia haitanensis is an economically important marine crop in southern China. The research on the life history of P. haitanensis is the basis of artificial cultivation, genetic breeding and other related research, but there is still a lack of systematic analysis of the whole growth and development process of P. haitanensis. The regulatory mechanism of P. haitanensis in its growth and development was revealed by comparing the transcriptome: photosynthesis is used to accumulate organic matter for the differentiation of Double Spore and Conchospore. Mismatch repair related genes are involved in the meiosis process of Conchospore; proline, actin and ubiquitin-binding enzymes increase Conchospore adversely to water. The process of Thallus cells into germ cells, photosynthesis, basal metabolic levels are reduced, DNA replication and chromatin remodeling processes are related to increased metabolism, and protein turnover is accelerated. The cytoskeleton, macromolecular HSP and phytohormone not only promote the formation of new vegetative algae, but also improve the adaptability of algae to the water environment. In addition. the phosphatidylinositol signaling system is an important signal for the adaptation of the laver filaments to different growth environments. These results provide a basis for the study of the life history of the P.haitanensis and the breeding of new varieties.
