一株鲤春病毒血症病毒全基因组序列分析

【摘要】 鲤春病毒血症(Spring Virernia Of Carp,SVC)是一种高致死性鲤科鱼类病毒病。课题组在2016年水生动物疫病监测期间,从我国北方地区某鲤鱼养殖场分离获得一株鲤春病毒血症病毒(Spring Virernia Of Carp Virus,SVCV),命名为SVCV-shlj4。全基因组序列分析显示该毒株基因组全长为11029 bp,从3′端到5′端包含5个开放阅读框,分别编码核蛋白(N,60-1394 nt,418 aa)、磷蛋白(P,1397-2363 nt,309 aa)、基质蛋白(M,2366-3081nt,223 aa)、糖蛋白(G,3084-4681 nt,509 aa)和RNA聚合酶(L,4686-11010 nt,2095aa),3′非编码区为59 bp,5′非编码区为19 bp。利用全基因组序列对SVCV-shlj4进行系统发育分析,结果显示目前世界范围内的SVCV分离株可以分为Ia、Ib、Ic、Id四个基因型,shlj4与我国现有的其余9株SVCV分离株聚为一簇,均为Ia基因型,其中SVCV-shlj4与我国SVCV A2(DQ491000.1,广东)基因组相似性最高(99.0%),与我国SVCV SH140501(KR012465.1,上海)相似性最低(97.4%)。本研究表明我国现行SVCV分离株均为Ia基因型,且基因组相似性高达97.4%以上。

【Abstract】 Spring virernia of carp(SVC) is a serious disease of common carp(Cyprinus carpio) with high mortality. In this study, a spring virernia of carp virus(SVCV)(named SVCV-shlj4) was isolated from a carp farms during the surveillance of aquatic animal disease in northeast China in 2016. Genome sequence analysis showed that the complete genome of SVCV-shj4 strain was about 11029 bp in length, and contained 5 open reading frames from the 3’ end to the 5’ end, encoding nuclear proteins(N, 60-1394 nt, 418 aa) and phosphoprotein( P, 1397-2363 nt, 309 aa), matrix protein(M, 2366-3081 nt, 223 aa), glycoprotein(G, 3084-4681 nt, 509 aa) and RNA polymerase(L, 4686-11010 nt, 2095 aa). The 3’ non-coding region is 59 bp, and the 5’ non-coding region is 19 bp. The phylogenetic analysis showed that the worldwide SVCV strains can be divided into four genotypes: Ia, Ib, Ic, and Id. Shlj4 and other 9 Chinese SVCV strains were clustered within the Ia genotypes. Among them, SVCV-shlj4 had the highest genome similarity(99.0%) with that of SVCV A2(DQ491000.1, Guangdong), and had the lowest genome similarity(97.4%) with SVCV SH140501(KR012465.1, Shanghai). This study suggests that the current Chinese SVCV strains are Ia genotypes, and the similarity is more than 97.4%.
