翘嘴鲌(Erythroculter ilishaeformis)精子生理特性及超低温冷冻的研究

【摘要】 采用实验生态—显微观察方法对翘嘴鲌精子的生理特性进行了研究,并用两步降温法超低温冷冻保存翘嘴鲌精子,筛选出了适宜的稀释液与抗冻剂配方。结果表明,翘嘴鲌适宜的盐度范围为0-4,pH为5.5-8时,精子激活率较高,其中pH为5.5时,精子运动时间与寿命最长;翘嘴鲌精子在4℃条件下保存84h仍有48.33%的激活率,在25℃条件下保存84h时精子死亡。以D-15为稀释液,10%乙二醇(EG)为抗冻剂,采用两步降温法超低温冷冻保存翘嘴鲌精子,解冻后精子活力最高,冻精激活率、运动时间和寿命分别为62.67%、63.33min和83.67min。

【Abstract】 Experimental ecology and microscopic techniques were investigated in order to understand the physiological characteristics of sperm of Erythroculter ilishaeformis.A two-step cooling procedure was employed to cryopreserve the sperm of Erythroculter ilishaeformis to selected effective dilutions and cryoprotectants.Results showed that the sperm motility was better when activated in solutions with a salinity of 0-4,the appropriate pH for better sperm activation rate was 5.5-8,for better sperm moving time and lifespan were 5.5.When the sperm were laid at4 ℃ in 84 h,they still had a 48.33%activation rate;when the sperm were laid at 25 ℃ in 84 h,they were dead.We employed a two-step cooling procedure to cryopreserve Erythroculter ilishaeformis sperm,the best frozen sperm motility appeared when the D-15 was the diluent,the10%ethylene glycol was antifreezer.The activation rate,moving time and lifespan of frozen sperm were 62.67%,63.33 min and 83.67 min,respectively.
