利用~1H-NMR代谢组学方法分析甲氨蝶呤对CIA大鼠的治疗效应

【摘要】 目的:利用~1H-NMR代谢组学方法研究与甲氨蝶呤治疗CIA大鼠作用相关的小分子代谢物。方法:建立胶原诱导关节炎大鼠模型,给予0.01mg/kg剂量的MTX灌胃治疗4周,对照组则给予生理盐水,根据大鼠的关节炎症状和病理特征对治疗效果进行评价。将治疗后收集的大鼠尿标本利用~1H-NMR进行检测。利用相关软件以及多元统计方法分析所获得的波谱图形,寻找与治疗相关的内源性代谢物。结果:8种小分子代谢物与治疗作用密切相关,这些代谢物与核酸代谢、能量代谢、半胱氨酸代谢等代谢途径有关。结论:~1H-NMR代谢组学方法可以分析甲氨蝶呤治疗CIA大鼠引起的代谢变化,治疗效果与多种代谢途径相关。

【Abstract】 Object.To investigate metabolic biomarkers related to therapeutic effects of methotrexate(MTX) in collagen-induced arthritis(CIA) rats using ~1 H-nuclear magnetic resonance(~1 H-NMR) based urine metabolomic analysis.Methods.CIA rats developed rapidly after immunized with collagen and treated by MTX 0.1 mg/kg for 4 weeks.The clinical signs and the histopathological features of CIA were obalyses were carried out to recognize the spectral pattern ofserved to evaluate the therapeutic effects.Collected urine samples were analyzed using 600 M~1 H NMR for spectral binning after treatment.The urine spectras were assigned by software and multivariate data an endogenous metabolites related to the spectra.Results.Eight endogenous metabolites were selected as putative biomarkers for predicting therapeutic response of MTX,and these metabolites were related to nucleic acid metabolism,energy metabolism,homocysteine metabolism and so on.Conclusions.The therapeutic effects of MTX can be predicted using ~1 H NMR based metabolomic study,and several metabolites may relat to the therapeutic action.
