补肾活血汤对透明带3多肽诱导的免疫性卵巢早衰小鼠模型血清IL-10、IFN-γ表达的影响

【摘要】 目的观察补肾活血对透明带3多肽(p ZP3)诱导的免疫性卵巢早衰(POF)模型小鼠血清IL-10、IFN-γ表达的影响,为探讨免疫性卵巢早衰患者相关治疗方案提供实验依据。方法取6~8周龄的健康雌性B6AF1小鼠75只,随机分为对照组和模型组和补肾活血汤组,每组25只。除对照组外,其余两组均采用透明带3多肽诱导造模,通过阴道脱落细胞涂片、血清性激素、卵巢组织形态及ZP3抗体免疫荧光染色鉴定建模成功。补肾活血汤组造模成功后予药物灌胃治疗28 d。给药结束后,酶联免疫吸附(Elisa)法检测各组小鼠血清IL-10、IFN-γ水平。结果 4周后90%以上小鼠造模成功。模型组小鼠外周血IL-10、IFN-γ表达水平上升,经本实验中补肾活血汤治疗后,外周血IL-10、IFN-γ水平下降。结论补肾活血汤可能过调节血清炎症因子水平,减轻透明带3多肽(p ZP3)诱导的炎症反应程度,从而改善免疫性卵巢早衰。

【Abstract】 Objective To observe the effect of Bushen Huoxue Decoction on serum IL-10, IFN-γ level in mice with autoimmune premature ovarian failure(POF) induced by zona pellucida 3 peptide(pZ P3).Methods The mice were randomly divided into three groups: control group(n=25), model group(n=25) and BSHXF-treated group(n=25). Autoimmune POF models were established in 50 female mice by immunization with pZ P3 and another 25 mice served as the control group. Mice were sacrificed 14 days after immunization. The POF models were verified by vaginalcytology, serum sex hormones,ovary histomorphology and ZP3 antibody immunohistochemistry. In the study, the mice in the BSHXF-treated group were given oral suspensions of 31.53 g/kg BSHXF once a day for 28 days. The serum IL-10, IFN-γ level of the mice were detected. Results Autoimmune POF models were estblished successfully more than 90% of the mice at 4 weeks after the immunization. Compared with those in the control group, the mice in the model group showed a smaller volume of the ovary, the atretic follicle of ovary was much in amount. Immunohistochemical studies showed that the model mice had redundant lymphocytes infiltrating around follicle compared to control mice.Compared to control group mice, the levels of IL-10 and IFN-γ in the serum of model mice significantly increased. But, compared to model mice, all the inflammation-related cytokine levels significantly decreased in BSHXF-treated mice. Conclusion Bushen Huoxue Decoction may regulate the level of serum inflammatory factors and reduce the extent of inflammatory response induced by the p ZP3, so as to improve immune premature ovarian failure.
