海洋小单孢菌FIM02523产Rakicidin B发酵工艺初步研究

【摘要】 目的提高海洋小单孢菌FIM02523发酵液中的生物活性成分Rakicidin B含量。方法采用单因素实验、正交设计实验等方法优化发酵培养基组成比例及培养条件。结果确定了最优Rakicidin B发酵培养基(%):可溶性淀粉4%,蔗糖1%,大豆蛋白胨1%,酵母粉2%,甘氨酸0.1%,NaCl 0.5%,MgSO4 0.05%,CaCO3 0.5%;最优摇瓶发酵条件:发酵周期为120小时,接种量5%,500ml摇瓶装量100ml,摇床转速220rpm,发酵温度30℃,培养基原始pH值为7.5。优化后发酵工艺Rakicidin B的发酵效价较初始工艺提高了约8倍。
【Abstract】 Objective To enhance the productivity of Rakicidin B by marine Micromonospora sp.FIM02523.Methods The fermentation medium and fermentation conditions were tested by single-factor experiment and Orthogonal experimental design.Results The optimum culture determined was as follows:Soluble starch 4%,sucrose 1%,soybean peptone 1%,yeast powder 2%,Glycine 0.1%,NaCl 0.5%,MgSO4 0.05%,CaCO3 0.5%;The optimum fermentation conditions were ascertained:fermentation time 120 hours,inoculation 5%,80 mL medium in 500 mL flask,rotation speed 220 r/min,culture temperature 30℃,pH 7.5 before sterilization.The last fermentation titer was improved approximately 8.3 times than that of the original.
