饥饿对鲤鱼脂肪酶和脂蛋白脂肪酶活性的影响

【摘要】 脂蛋白脂肪酶(LPL)是脂肪代谢的关键酶之一,本实验采用对硝基苯酚法测定了鲤鱼脂肪酶及LPL酶在正常摄食和饥饿状态下各组织脂蛋白脂肪酶的酶活。结果表明正常摄食的鲤鱼血清、肌肉、脂肪、肝、肠和心总脂肪酶比活力分别为531.7±22.1 U/g、213.1±87.0U/g、198.7±123.6U/g、151.7±11.6 U/g、998.4±398.4U/g、763.3±122.2U/g,饥饿状态下各组织脂肪酶比活力分别为118.5±15.6 U/g、99.5±19.6 U/g、120.4±39.3U/g、276.3±U26.4/g、1600.0±567.8U/g和481.8±56.9U/g,与正常摄食相比饥饿状态下鲤鱼肝脏脂肪酶比活力显著升高(P<0.05);使用1M Nacl抑制LPL酶活,测出正常摄食状态下鲤鱼各组织的LPL比活力分别为340.1±110.5 U/g、132.5±67.2 U/g、115.5±32.6 U/g、141.7±8.4 U/g、337.3±38.1 U/g、264.5±44.9 U/g,饥饿下则为41.7±12.8 U/g、91.0±20.9 U/g、74.5±37.2U/g、226.9±24.8U/g、378.8±295.7 U/g和337.4±40.8 U/g,与正常摄食相比饥饿状态下鲤鱼肝脏中脂蛋白脂肪酶酶活力显著升高(P<0.05)。测定鲤鱼脂肪酶和脂蛋白脂肪酶活性,为以后研究各组织和器官的脂蛋白脂肪酶表达研究奠定了基础。

【Abstract】 Lipoprotein lipase(LPL) is one of the key enzymes of fat metabolism. In this experiment, the activity of lipase and LPL enzyme in normal feeding and starvation were measured by p-nitrophenol method. The results showed that the lipase activities of serum, muscle, fat, liver, intestine and heart in normal feeding Cyprinus carpio were 531.7 ± 22.1 U/g, 213.1 ± 87.0 U/g, 198.7 ± 123.6 U/g, 151.7±11.6 U/g, 998.4 ± 398.4 U/g, 763.3 ± 122.2 U/g, respectively. The activity of lipase in hungry state was 118.5 ± 15.6 U/g, 99.5 ± 19.6 U/g, 120.4 ± 39.3 U/g, 276.3 ±26.4 U/g, 1600.0±567.8 U/g and 481.8 ± 56.9 U/g, respectively. Compared with normal diet, the activity of lipase in Cyprinus carpio was significantly increased(P <0.05). The use of 1 M Nacl inhibited LPL activity, the LPL activity of the tissues was 340.1 ± 110.5 U/g, 132.5 ± 67.2 U/g, 115.5 ± 32.6 U/g, 141.7 ± 8.4 U/g, respectively. Under normal feeding status the LPL activity of tissues in Cyprinus carpio was 337.3 ± 38.1 U/g, 264.5 ± 44.9 U/g, 41.7 ± 12.8 U/g, 91.0 ± 20.9 U/g, 74.5 ± 37.2 U/g, 226.9 ± 24.8 U/g, 378.8 ± 295.7 U/g and 337.4 ± 40.8 U/g, and compared with the normal diet the activity of lipoprotein lipase in the liver were significantly increased(P <0.05). The lipase activity of lipase and lipoprotein in carp were determined, which laid a foundation for the study of lipoprotein lipase expression in tissues and organs.
