1高温干旱环境下考来木幼苗的活性氧代谢

ROS Metabolism of Correa Carmen Seedlings under Combined Elevated Temperature and Drought Stress

【摘要】 以盆栽二年生考来木幼苗为试材,在人工气候室中模拟高温、干旱环境,研究在高温、干旱复合胁迫下考来木的活性氧代谢。结果如下:(1)干旱或高温单一胁迫下,考来木幼苗的H202、等活性氧代谢物含量及MDA含量增加,SOD酶活性亦同步显著增高。(2)在高温、干旱复合胁迫下,活性氧代谢物含量及抗氧化性酶活性均显著高于单一胁迫,SOD活性近乎叠加增长;较之单一胁迫,复合胁迫下MDA含量并无显著升高。(3)随胁迫时间延长或胁迫程度增加,抗氧化性酶活性持续增强;当胁迫增加到一定程度或持续一定时间后,抗氧化酶活性显著下降。(4)干旱胁迫对考来木的伤害大于高温胁迫。结果表明,考来木可通过大幅提高抗氧化性酶系统的活性来抵御高温干旱胁迫所产生的活性氧伤害,因而具有较强的耐高温干旱能力。但抗氧化酶活性的提高是有限的,高强度或长时间胁迫下活性氧大量积累,抗氧化性酶被破坏而活性下降,则作物受害。本研究为考来木的栽培与推广、抗逆品种的培育等提供科学依据。

【Abstract】 Cultivated Correa carmen seedlings in pot as test materials in phytotron to understand the physiological mechanism that Correa carmen seedling adapt to combined stress of elevated temperature and drought.The air temperature in phytotrons(T) were set up 3 gradients to simulate high temperature stress:25℃,35℃,45℃.The water content(S) in pot soil were also set up 3 gradients to simulate different degree of drought:S1:90% of field water holding capacity,S2:60% of field water holding capacity,S3:30% of field water holding capacity,each treatment repeated 3 times.Gathered functional leaves respectively from each pot to measure the reactive oxygen metabolites content,MDA content and the activities of antioxidant enzyme on 0 day,7 th day,14 th day,21 th day,28 th day.The results showed that:(1)Under the single stress of drought or high temperature,H2O2 contents,rates of production and MDA contents increased significantly in functional leaves of Correacarmen seedlings,at the same time the activities of SOD,POD and CAT antioxidant enzyme also increased significantly.(2)Under the composite stress of high temperature and drought,ROS content and antioxidant enzyme activities were significantly higher than that of single stress,SOD activity nearly superposition growth;Compared with the single stress,MDA content had no significant change under complex stress.(3) ROS content and antioxidant enzyme activities progressively increased along with the extension of stress time and the increase of the stress degree.Nevertheless,antioxidant enzyme activities decreased significantly when the stress increased to a certain degree or lasted a certain period.(4)The ability of resistance to high temperature stress of Correa carmen seedlings was more stronger than that to drought.All above results showed that Correa carmen seedlings could significantly improve its activity of antioxidant enzyme system to defense ROS caused by high temperature and drought stress,thus they had high ability of resistance to high temperature and drought.Nevertheless,it was limited that the degree of increase of the antioxidant enzymes activity,at the same time plants hurt by ROS finally.This study provided the scientific basis for Correa carmen cultivation and promotion,and the materials to breed garden plant varieties with resistance to stress of elevated temperature and drought.
