包埋辅酶Q10的纳米结构脂质载体的制备及性能研究

【摘要】 本文采用高压均质法制备了运载辅酶Q10的纳米结构脂质载体(CoQ10-NLC),通过粒径、电位、包封率等测试手段对其制备条件进行优化,通过差示扫描热量仪(DSC)和透射电镜(TEM)对其结构和形貌进行表征,并通过体外模拟透皮实验考察了CoQ10-NLC的皮肤吸收效果。结果表明,CoQ10-NLC的最佳制备条件为:固液脂质的质量比为1:1,表面活性剂的质量分数为1.5%,高压均质压力为1000bar,循环次数为7次,并且制得的CoQ10-NLC能够增加CoQ10在皮肤中的渗透量。
【Abstract】 In this study,CoQ10-loaded nanostructured lipid carriers(CoQ10-NLC) were prepared by high pressure homogenization method.The formulations were optimized by the determination of particle size,Zeta potential,encapsulation efficiency,ect.The structure and morphology of CoQ10-NLC were characterized by differential scanning calorimetry(DSC) and transmission electron microscopy(TEM).The skin absorption effect of CoQ10-NLC was assessed by in-vitro transdermal study.The optimized preparation conditions were that the mass ratio of solid and liquid lipid was 1:1,the mass fraction of surfactant was 1.5%,the preparation pressure was 1000 bar,the cycles were 7 times,and in-vitro transdermal study indicated CoQ10-NLC enhanced the permeation of CoQ10 in skin.
