Ultrastructure,Morphological and Molecular Characterization of New and New Recoreded Species of Hemicriconemoides from China

【摘要】 Sheathoid nematodes of the genus Hemicriconemoides are migratory root-ectoparasites of many plants including various agricultural crops and fruit trees.They are generally found inhabiting tropical and subtropical areas of the world and have been reported from Africa,America,Asia,Australia and southern Europe,presently genus consists of 52 valid species.In this study we provide morphological and molecular characterization of one new and one new recorded species viz.Hemicriconemoides n.sp.and H.kanayanesis isolated from Metasequoia glyptostroboides and Camelliae uruku in Hangzhou,Zhejiang,respectively.The new species is most similar to H.californianus and considered as a cryptic species.Morphologically the new species can be characterized by cuticular sheath well separated at anterior region,cuticle smooth with retrorse annuli,lip region slightly set off and roughly square in outline.Stylet7383μm long,excretory pore 45 annuli posterior to pharyngeal bulb,vulva posteriorly located88% 91%of the total body,tail conical with bluntly pointed terminus.H.kanayaensis can be characterized by its exclusive lip morphology(set off,marked with constriction),cuticular sheath attached at anterior and vulval region,stylet 6678μm long,excretory pore located 4 5 annuli posterior to pharyngeal bulb,vulva posteriorly located 88%90%of the body,tail elongated widely conoid,ended up in a rounded terminus.Scanning electron microscopic observations and molecular characterization based on COI of mtDNA,18 S,D2-D3 28 S and ITS of rDNA sequences for both species also provided.To date sixteen Hemicriconemoides species have been reported from China,where H.kanayaensis is the 17th recorded species.Our results suggested that the concomitant use of morphological and molecular characterization help to remove the taxonomic opaqueness and paucity of knowledge regarding species identity.This will help to improve its relevance for posterity and will further elucidate the deeper levels of phylogenetic relationships,among species of genus Hemicricortemoides.
