功能化硼酸磁性纳米材料用于生物样品中糖蛋白/糖肽的富集

【摘要】 蛋白质糖基化是生物体内普遍存在和复杂的翻译后修饰过程,比如受体激活、细胞的信号传导、免疫应答等生物过程[1]。然而,在复杂生物体系中,许多糖蛋白的含量都相对较低,且被高丰度的非糖蛋白所干扰,这使得应用质谱对其进行检测很困难[2]。因此,从复杂生物体系中选择性地分离和富集糖蛋白依然具有挑战。到目前为止,凝集素亲和技术、肼腙化学反应法、亲水作用法等广泛应用于糖蛋白的富集。因为硼酸可以可逆地与顺式二醇类分子结合并且容易富集,硼酸亲和材料已经用于了糖蛋白的分离富集[3]。本工作中我们采用蒸馏沉淀聚合法、原子转移自由基聚合法、巯基-烯(炔)点击化学反应方法制备硼酸配体修饰的磁性Fe3O4和石墨烯材料,并将其成功地用于生物样品中糖蛋白/糖肽的分离富集。

【Abstract】 Protein glycosylation is one of the most important posttranslational modifications and plays a significant role in cell attachment-recognition,cell transportation,immune response,and other areas.However,the detection of glycoproteins is severely interferred by other non-glycoproteins due to the inherent low abundance of glycoproteins in mass spectrometry analysis.Therefore,efficient enrichment/separation of glycoproteins from the complex biological samples is challenges.In the past decade,several strategies including lectin-affinity chromatography,hydrazide chemistry,and hydrophilic interaction chromatography have been developed for separation of glycoproteins and glycoforms.Alternatively,boronic acid-based methods have gained increasing popularity due to boronic acids can covalently form cyclic esters with the cis-diols moiety of biomolecules in a basic aqueous media,while the reversible release can be manipulated by switching to acidic conditions.Herein,we describe some efficient methods including distillation precipitation polymerization(DPP),atom transfer radical polymerization and thiol-ene(TE)/thiol-yne(TY) click chemistry to prepare the boronate functionalized Fe3O4 or graphene to enrich glycoproteins/glycopeptides from biological samples.
