偷死野田村病毒在脊尾白虾中的垂直传播

【摘要】 脊尾白虾是虾类养殖池塘中常见的甲壳动物,在河口、近海等野外水体中也广泛存在,它在偷死野田村病毒(Covert mortality nodavirus,CMNV)的广泛传播中可能起到了中间宿主和寄存宿主的作用。为筛查CMNV的传播途径,本文对CMNV在脊尾白虾中垂直传播的可能性进行了研究,利用RT-LAMP、nRT-PCR、ISH和TEM对染病亲体的卵巢、精巢、受精卵和后代幼体进行了检测分析。RT-LAMP和nRT-PCR结果显示染病脊尾白虾亲体的卵巢组织、精巢组织均呈现CMNV阳性;感染CMNV的雌性个体和健康雄性个体交尾所产生的受精卵、健康雌性个体和感染CMNV的雄性个体交尾所产生的受精卵也均呈现CMNV阳性。ISH结果显示,卵细胞、孵化过程中的幼体细胞内均出现较强的阳性信号。TEM结果表明,感染CMNV脊尾白虾亲体的卵原细胞、卵细胞、精细胞、受精卵和孵化中的幼体细胞内均可见大量的CMNV样颗粒。综合上述结果,说明CMNV可通过精细胞和卵细胞在脊尾白虾中进行垂直传播。本研究结果也提示,CMNV在脊尾白虾中垂直传播的风险值得高度关注。

【Abstract】 Viral covert mortality disease(VCMD) caused serious loss of shrimp aquaculture in China in recent years and the Exopalaemoncarinicauda was suspected to be one important factor in perpetuating the high prevalence of the covert mortality nodavirus(CMNV) infections for its perennial presence in the farming ponds and wild water. Experiments were carried out to determine the possibility of vertical transmission of CMNV in this study. CMNV infection in ovarian tissue, testis tissue, fertilized eggs, and larvae of the artificial infected E.carinicauda were investigated by using methods of RT-LAMP, nRT-PCR, ISH and TEM. The ovarian and testis of artificial infected parental E.carinicauda were proved to be positive of CMNV by RT-LAMP and nRT-PCR. Fertilized eggs were also founded to be CMNV positive of nRT-PCRwhether the fertilized eggs were originated fromthe CMNV positive female broodstock mating with the CMNV negative male broodstock, orthey were originated fromthe CMNV negative female broodstock mating with the CMNV positive male. The result of ISH indicated that the positive signals were evident in theoocytesand larvae. By TEM analysis, CMNV virions were observed in oogoniums, oocytes, spermatocytes, fertilized eggs and larvae.
