尼古丁对大鼠牙槽骨OPG、RANKL基因表达的影响
The effect of nicotine on OPG and RANKL gene expression in the alveolar bone of rats

【摘要】 目的:通过对大鼠腹腔注射不同剂量的尼古丁,建立动物模型。观察大鼠牙周组织的变化、破骨细胞数目和牙槽骨OPG、RANKL基因表达的变化,探讨尼古丁对大鼠牙槽骨骨吸收的影响。方法:选取SPF清洁级SD雄性大鼠21只,随机分为对照组、低剂量尼古丁组、高剂量尼古丁组,分别腹腔注射生理盐水、0.8mg/kg尼古丁溶液、3.2mg/kg尼古丁溶液。3个月后处死大鼠,收集左下牙槽骨制作牙体牙周联合切片,HE染色观察大鼠牙周组织的变化,Trap染色后进行牙槽骨表面破骨细胞的计数,免疫组化染色检测大鼠牙槽骨中OPG、RANKL蛋白的表达,收集右下牙槽骨,q PCR技术检测大鼠牙槽骨中OPG、RANKL mRNA的表达。结果:低剂量尼古丁组和高剂量尼古丁组牙周纤维排列较对照组紊乱,牙槽骨表面形态不规则。对照组、低剂量尼古丁组、高剂量尼古丁组破骨细胞数目依次增多,两两比较,差异具有统计学意义(P<0.05)。低剂量尼古丁组、高剂量尼古丁组OPG表达阳性细胞数目较对照组少,颜色较对照组浅,RANKL表达阳性细胞数目较对照组多,颜色较对照组深;对照组、低剂量尼古丁组、高剂量尼古丁组OPG mRNA的相对表达量依次减少,RANKL mRNA的相对表达量及RANKL/OPG比值依次增高,两两比较,除高剂量尼古丁组与低剂量尼古丁组的OPG mRNA表达无明显差异外(P>0.05),其余各组比较均有统计学意义(P<0.05)。结论:尼古丁可能通过下调牙槽骨OPG基因的表达,上调RANKL基因的表达,调节破骨细胞的数目,影响牙槽骨的骨吸收,但具体机制有待进一步的研究。
【Abstract】 Objective: Via the injection of different dose of nicotine into SD rats through abdomen, the animal model was established. The changes of the periodontal tissue, the quantity of osteoclast and the OPG, RANKL gene expression in the alveolar bone were observed, the effect of nicotine on alveolar bone resorption in rats was explored. Methods: 21 male SD rats were randomly divided into three groups: control group, low dose nicotine group and high dose nicotine group respectively. The animals were injected into saline solution, 0.8 kg/kg nicotine solution and 3.2 kg/kg nicotine solution respectively through abdomen. The rats were sacrificed after 3 months, and the left lower alveolar bone was made into slices. The histological changes of the periodontal tissue was observed by HE staining, the quantity of octeoclast was recorded after trap staining, and the expression of OPG, and RANKL protein was observed by immunohistochemistry staining. The right lower alveolar bone was used to detect the mRNA of OPG and RANKL expression by q PCR technique. Results:Periodontalfibers arranged irregularly, alveolar margin was irregular in the low dose nicotine group and the high dose nicotine group compared with that in the control group. The quantity of osteoclast increased in the control group, low dose nicotine group, and high dose nicotine group in turn, pairwise outcome, there were significant difference among these three groups(P<0.05). The number of OPG positive cells was less in the low dose nicotine group and high dose nicotine group compared with the control group, and the color is less deep; the quantity of RANKL positive cells in the low dose nicotine group and the high dose nicotine group was more than that in the control group. The expression of OPG mRNA in the control group, low dose nicotine group and high dose nicotine group decrease in turn, and the expression of RANKL mRNA and the ratio of RANKL to OPG in the control group, low dose nicotine group and high dose nicotine group increased in turn, obvious difference could be seen between each two groups(P<0.05), except that the expression of OPG mRNA in the high dose nicotine group and that in low dose nicotine group had no statistic significant(P>0.05). Conclusion:Nicotine may down regulate the expression of OPG gene and up regulate the expression of RANKL gene in the alveolar bone, as a result, regulating the number of osteoclasts, therefore the alveolar bone resorption was affected. While the mechanisms of nicotine on the alveolar bone resorption is going to research in the future.
