水老化处理对DMADDM改性玻璃离子水门汀抗菌作用的影响

【摘要】 目的:在前期研究中我们将季铵盐单体二甲氨基十二烷基甲基丙烯酸脂(dimethylaminododecyl methacrylate,DMADDM)添加至传统玻璃离子水门汀进行抗菌改性,证实了DMADDM改性的玻璃离子水门汀具有较强的抗菌性能,可以有效抑制口内原位生物膜活性。本研究旨在探讨水老化处理对DMADDM改性玻璃离子水门汀抗菌性能的影响。方法:将不同质量分数(1.1%,2.2%)的DMADDM添加到玻璃离子中制备抗菌改性的玻璃离子作为实验组(n=216),未添加抗菌单体的玻璃离子作为对照组(n=108)。样本分别浸入37℃无菌去离子水中进行1天和6个月的水老化处理,采用口内原位生物膜模型评价样本对菌斑生物膜形成及活性的影响。玻璃离子样本固定在个体定制的口内保持器上,由六名健康的志愿者分别佩戴保持器24h、48h,通过荧光显微镜和扫描电镜观察分析实验材料表面不同时段口内生物膜形成情况,评价水老化处理的材料对抗口内生物膜作用。结果:材料表面的生物膜活性随DMADDM的增加显著降低,荧光显微镜结果显示DMADDM抗菌改性的实验组材料表面生物膜活性明显低于对照组,水老化处理1天和6个月的样本表面生物膜形成情况无显著差异。结论:经过水老化处理的DMADDM抗菌改性的新型玻璃离子水门汀材料仍具有持久的抗口内原位生物膜活性,是一种有临床应用潜力的新型防龋材料。

【Abstract】 Objective: In the previous study, we developed a new antibacterial GIC containing a dimethylaminododecyl methacrylate(DMADDM). It was proved that the DMADDM modified GIC greatly inhibited biofilm in situ. The aims of this study were to investigate the effect of water-ageing for 6 months on antibacterial properties of GIC incorporated with DMADDM. Methods: Biofilms were allowed to form in situ on GIC specimens(n=216) which contained DMADDM(1.1 wt.% or 2.2 wt.%). Samples without DMADDM served as control(n=108). Specimens were water-aged for 1 d and 6 months at 37℃, then fixed on custom made splints and exposed to the oral cavity in six healthy volunteers for 24 and 48 h, respectively. The bacterial vitality of biofilm was analyzed by fluorescence microscopy(FM). The bacterial morphology and biofilm accumulation were determined by scanning electron microscopy(SEM). Results: Increasing DMADDM mass fraction monotonically reduced the biofilm viability. FM analysis revealed that bacterial vitality of the biofilm on both DMADDM concentration samples was significantly lower than controls after 24 hours as well as after 48 hours in situ. DMADDM modified GIC exhibited a long-term antibacterial performance, with no significant difference between 1 d and 6 months.Conclusions: Incorporating DMADDM in GIC yielded potent and long-lasting antibacterial properties. The experimental material is promising to inhibit caries.
