芍药扦插生根过程中POD、PPO和IAAO活性变化及生根部位形态解剖研究

【摘要】 以芍药品种‘大富贵’‘紫凤羽’‘奇花露霜’和‘桃花飞雪’为试验材料,通过扦插试验,比较了不同品种芍药扦插生根的差异;插穗生根过程中POD、PPO和LAAO的活性变化,采用石蜡切片法研究了不定根发生的组织解剖学特性。结果表明:4个芍药品种中,‘大富贵’生根率最高可达73.33%、生根数量和根长都优于其他3个品种,扦插效果最好,其次分别是‘紫凤羽’‘奇花露霜’和‘桃花飞雪’;生根过程中POD活性和PPO活性程"上升-下降"的单峰变化,二者在扦插后第40d活性最高且与生根时间相吻合,IAAO活性呈下降趋势,在扦插后40d生根时达到最低值;芍药茎段中本身并不存在潜伏根原基,属于诱导生根类型;芍药插穗不定根的发生部位位于维管形成层,属于皮部生根类型。

【Abstract】 The rooting method of cuttings of Paeonia lactiflora were studied with ’ Da fugui ’,’ Zi fengyu ’,’ Qi hualushuang ’ and ’ Tao huafeixue ’ by cuttings.The different performances between various cultivars were compared.The changes of three kinds of enzyme activities(POD,PPO and IAAO) during the roots formation were studied either.The anatomical study on the occurrence of adventitious roots were carried out using paraffin section method.The results showed that:The results of cuttings used 10 mg/L IAA treatments was the best,the rate of rooting was 73.33%.Among these 4 cultivars,the rooting rate,rooting numbers and roots ’ length exhibited the best,followed by ’ Zi fengyu ’,’ Qi hualushuang ’ and ’ Yang feichuyu ’.The POD and PPO activity showed an "increase and decrease" unimodal change trend,the highest point was at the 40 days after cutting which match the time of rooting.The IAAO activity decreased and reached the lowest point at the 40 days after cutting.There were not any root anlagen before cutting in Paeonia lactiflora.The adventitious roots mainly emerged in the cortex and origined from the parenchyma cells between xylem and pith.
