豆瓣绿胚性细胞悬浮培养研究

【摘要】 本研究以豆瓣绿无菌苗叶片为外植体诱导获得质地疏松、稳定性高的颗粒状胚性愈伤组织,开展胚性细胞悬浮培养研究。结果表明:豆瓣绿胚性愈伤组织最适继代培养基为MS+30g/L蔗糖+琼脂6g/L+6-BA1.0mg/L+2,4-D 1.0mg/L,继代间隔14d;体细胞悬浮最佳培养基为MS+30g/L蔗糖+6-BA 1.0mg/L+NAA0.5mg/L,继代间隔7d;体细胞悬浮最佳碳源为蔗糖,蔗糖最佳浓度为30g/L;体细胞悬浮摇床最佳转速为110r/min。本研究考察激素种类、激素浓度、碳源种类、糖浓度、摇床转速对体细胞胚悬浮体系的影响,并采用解剖镜进行形态学观察,为后期研究体细胞胚胎形态结构奠定基础。

【Abstract】 In this study,the embryogenic callus with high density and high stability was induced by the leaves of Peperomia obtusifolia as explants,and the embryogenic cell suspension culture was studied.The results showed that the optimal subculture medium was:MS + 30 g/L sucrose +agar 6 g/L+6-BA1.0 mg/L + 2,4-D 1.0 mg/L and subculture 14 d;The optimum medium for somatic cell suspension was MS +30 g/L sucrose + 6-BA 1.0 mg/L + NAA 0.5 mg/L,the next generation interval was 7 d;the optimal carbon source for somatic cell suspension was sucrose,the optimum concentration of sucrose was 30 g/L;somatic cell suspension The optimum speed of the shaker is 110 r/min.The effects of hormone type,hormone concentration,carbon source type,sugar concentration and shaking speed on the somatic cell suspension system were investigated.Morphological observation was carried out by using dissecting microscope to lay the foundation for the later study of somatic embryonic morphology.
