姜花中转录抑制因子HcJAZ1和HcJAZ2的克隆与功能信息学分析
Cloning and Functional Bioinformatics Analysis of HcJAZ1 and HcJAZ2 Gene from Hedychium coronarium

【摘要】 以白姜花(Hedychium coronarium)为材料,利用RT-PCR技术克隆得到两个JA途径的转录抑制因子JAZ(Jasmonate ZIM-domain)基因,HcJAZ1和HcJAZ2。HcJAZ1基因全长1238bp,具有完整的开放阅读框708bp,编码235个氨基酸,预测分子量为25.7kDa,等电点(pI)为9.26。HcJAZ2基因全长1505bp,具有完整的开放阅读框780bp,编码259个氨基酸,预测分子量为28.5kDa,等电点(pI)为7.62。蛋白家族结构域分析显示HcJAZ1和HcJAZ2蛋白序列均含有和JAZs形成同源或者异源二聚体相关的高度保守的TIFY(又称ZIM)结构域,以及和泛素化降解,核定位相关的CCT1(又称Jas)结构域。氨基酸同源序列比对结果表明不同的JAZ基因氨基酸序列的变化较小。聚类分析进化树结果表明,HcJAZ1和HcJAZ2与拟南芥的JAZ1和AtJAZ2亲缘关系相对最近,且具有较高的同源性。核定位信号分析预测结果显示HcJAZ1和HcJAZ2均在N端存在一段潜在核定位信号,NLS预测值分别为3.4和3.1。蛋白疏水性预测分析结果表明HcJAZ1,HcJAZ2均有较强的疏水性。二级结构预测分析结果显示HcJAZ1含有68.5%成α螺旋残基,37.9%成β折叠残基,15.7%成β转角残基;HcJAZ2含有56.9%成理螺旋残基,41.9%成β折叠残基,17.7%成β转角残基,这和AtJAZ1,AtJAZ2的二级结构表现出较高的相似性。此外,蛋白三维建模分析显示TIFY结构域和CCT1结构域的空间位置关系在HcJAZ1和HcJAZ2中较为相似。
【Abstract】 A transcription inhibitors gene,named HcJAZl and HcJAZ2,was isolated from the flowers of H.coronarium using the gene specific primers which obtained according to HcJAZ sequence by the method of RT-PCR technology.The HcJAZl gene was 1238 bp in length,containing a 708 bp ORF which encoded 235 amino acid residues of 25.7 kDa molecular weighth.The HcJAZ2 gene was 1505 bp in length,containing a 780 bp ORF which encoded 259 amino acid residues of 28.5 kDa molecular weighth.Protein predicted domain indicated that HcJAZl and HcJAZl have the typcial high conserved site TIFY domain which can form homodimer itself or heterodimers with other receptors and CCT_ 1 motif which can bind other transcripton factors including COIl.The result of amino acid sequence alignment reveals that the protein HcJAZl and HcJAZ2 poss some kind of the typical TIFY domain and CCT_ 1 motif of JAZ gene families compared to the JAZ in arabidopsis thaliana.Phylogenetic tree analysis for AtJAZl-12 as well as HcJAZl and HcJAZ2 indicates that AtJAZ1 and AtJAZ2 are the ones most closely related to HcJAZl and HcJAZ2.Subcellular localization shows the NLS scores of the HcJAZl and HcJAZ2 is 3.4 and 3.1,respectively.The prediction of the hydrophobic shows that HcJAZl and HcJAZ2 are high lyophobic.The secondary structure prediction results manifests that HcJAZl has 68.5%α-helix,37.9%p-sheet,15.7%β-turn;HcJAZ2 has 56.9%α-helix,41.9%13-sheet,17.7%p-turn,it is highly similar with what in AtJAZl and AtJAZ2.Besides,the results of the 3D structure prediction shows the similar special relationship between TIFY domain and CCT_ 1 domain in both HcJAZl and HcJAZ2.
