芍药与牡丹组间杂种‘Bartzella’的微繁殖技术研究

【摘要】 本文以芍药与牡丹组间杂种(伊藤杂种)‘Bartzella’的鳞芽为外植体,初步建立了其微繁殖体系。主要结果包括:①‘Bartzella’的最佳取材时间为3月下旬;②WPM培养基中Ca(NO32浓度提高至原浓度的4倍(2224mg·L-1),可显著促进组培苗的生长和增殖;添加有机物水解酪蛋白0.25g·L-1也可在一定程度上提高组培苗增殖率。③生根培养前,将无根苗先接种于培养基1/2 MS中复壮培养10天,然后转入1/2 MS+腐胺1.0mg·L-1+IBA 1.0mg·L-1培养基中诱导根原基,冷处理12天后转常温黑暗环境下诱导培养30天,最后在根形成培养基1/2MS+活性炭4.0g·L-1中培养20天,生根率可达75.51%。④生根质量对移栽成活率影响显著,根数多于3条且愈伤组织小的生根苗移栽成活率高达73.58%。

【Abstract】 The in-vitro cultured shoots of intersectional hybrids between herbaceous and tree peonies ’ Bartzella’ were used to develop a protocol for micro-propagation.The results showed that:①The optimal sampling time of ’ Bartzella’ was late March.②Modification of WPM basal medium on fourfold Ca(NO32 and adding 0.25 g · L-1 casein acid hydrolysate were effective on the improving of multiplication and growth of ’ Bartzella’.③For optimal rooting,the shoots were cultured on 1/2MS for 10 days,then cultured on 1/2 MS + putrescine 1.0mg · L-1 + IBA 1.0mg · L-1 for 30 days to induce rooting after a cold treatment(4℃)for 12 days,and then transferred onto root formation medium(1/2MS + Activated carbon 4.0mg · L-1)for20 days,so that 75.51%rooting rate was achieved finally.④The rooting quality had significantly effect on survival of in-vitro rooted plantlets in ’ Bartzella’.Plantlets with root number more than three and less callus survived 73.58%.
